- Establishment and genomic validation of novel patient-derived xenograft models for drug discovery in gastrointestinal stromal tumor. [Journal Article]BMC Cancer. 2026 Aug 10; 26(1).BC
- Most gastrointestinal stromal tumor (GIST) patients develop clinical resistance to approved tyrosine kinase inhibitors (TKI). The development of effective treatment strategies for refractory GIST requires identification of novel targets for investigational agents and well-characterized preclinical models, like patient-derived xenograft (PDX) models, to evaluate them. While GIST PDX models have be…

- Diagnostic interpretation of CFH expression in pan-cancer analyses: the importance of factor H-like 1 isoform resolution. [Letter]BMC Cancer. 2026 Oct 11; 26(1).BC
- Recent pan-cancer analyses have identified Complement Factor H (CFH) expression as an immune-related and prognostic biomarker. However, interpretation of CFH transcriptional data requires consideration of isoform specificity. The CFH gene encodes both full-length factor H (FH) and the biologically active splice isoform factor H-like 1 (FHL-1), which differ in structure and tissue localization. Co…

- A novel "early warning" system for biochemical recurrence in prostate cancer: development and validation of the SII-HALP combined score. [Journal Article]
- CONCLUSIONS: Based on internal validation, the preliminary SII-HALP combined score may serve as an exploratory, low-cost prognostic tool that contributes to optimizing risk stratification for 1-year biochemical recurrence. Reflecting a possible host-tumor biological interaction, it offers a conceptual pathway for individualized risk triage, pending further robust external validation and extended longitudinal follow-up.

- Despite improved overall stratification, the 9th edition stage IIIA category remains prognostically heterogeneous: insights from a T3N2a subgroup analysis. [Journal Article]
- CONCLUSIONS: While the 9th edition TNM system improves overall prognostic stratification for resected occult N2 NSCLC, the stage IIIA category remains prognostically heterogeneous. T3N2a patients have outcomes more consistent with IIIB than IIIA, suggesting that further refinement of stage grouping may be warranted.

- Impact of and recovery after Covid-19 on breast cancer diagnosis and care in Johannesburg, South Africa. [Journal Article]
- CONCLUSIONS: The already high prevalence of late-stage at diagnosis increased further during the COVID-19 pandemic with evidence of recovery at both sites in the post COVID-19 period. Treatment initiation delays increased and surgery volumes decreased during the pandemic and did not recover to pre-pandemic levels. Our study shows current cancer human and facility resources are insufficient to manage the steadily increasing BC burden in Johannesburg, South Africa.

- Co-mutation landscape and prognostic impact of genomic complexity in EGFR-mutant non-small cell lung cancer. [Journal Article]
- CONCLUSIONS: Our study demonstrates that prognosis in EGFR-mutant NSCLC is strongly influenced by broader genomic context, particularly TP53 mutation and overall co-mutation burden. EGFR subtype-specific survival differences and TP53/ATM-defined DDR-related findings should be interpreted cautiously because of study-origin heterogeneity, incomplete clinical annotation, and the limited DDR gene definition.

- Integrative transcriptomic and experimental analyses identifies SDF4 as an astrocyte-expressed regulator of glioma progression through PI3K-AKT signaling. [Journal Article]
- CONCLUSIONS: Our research underscores the critical role of SDF4 in glioma progression, highlighting its potential as a strategic target for glioma management and as a prognostic marker. Additionally, these findings pave the way for novel therapeutic strategies.

- Co-expression of non-signaling CARs enhances EGFRvIII-specific CAR T-cell cytotoxicity against head and neck squamous cell carcinoma. [Journal Article]
- CONCLUSIONS: EGFR nsCAR co-expression can provide a modular strategy to enhance EGFRvIII CAR T-cell cytotoxicity under conditions of limited target antigen availability. These findings provide an in vitro proof of concept for additional target engagement through a non-signaling receptor and warrant further evaluation in more complex preclinical models.

- Insights into the survival impact of delayed immunochemotherapy in newly diagnosed diffuse large B-cell lymphoma: results from a nationwide registry study. [Journal Article]
- CONCLUSIONS: Early treatment in DLBCL is primarily driven by adverse disease characteristics. A residual association between longer DTI and improved survival persisted after IPI adjustment, suggesting DTI may be an independent prognostic factor in less aggressive disease.

- Comparative effectiveness of neoadjuvant therapy combined with stent placement versus stent alone in obstructive colorectal cancer: a multicenter retrospective analysis. [Multicenter Study]
- CONCLUSIONS: SEMS-NAT for obstructive CRC may is an important factor for improved surgical quality and OS benefit in 2021-2025. This study provides crucial evidence for optimizing treatment strategy by recommending a surgery interval.

- The effect of adding ovarian function suppression to adjuvant endocrine therapy in premenopausal patients with hormone receptor-positive, HER2-positive breast cancer who have undergone neoadjuvant therapy: a single-center experience. [Journal Article]
- CONCLUSIONS: In this retrospective real-world study, no independent association between OFS and survival outcomes was observed in premenopausal patients with HR-positive/HER2-positive breast cancer treated with anti-HER2-based neoadjuvant therapy. Patients achieving pCR had excellent prognoses regardless of OFS use. Given the limited statistical power and the predominantly tamoxifen-treated cohort, these findings should be interpreted with caution. Prospective studies are warranted to better define the role of OFS in this patient population.

- Belantamab mafodotin plus venetoclax in t(11;14)-positive relapsed or refractory multiple myeloma - the BELI(E)VE trial. [Journal Article]
- CONCLUSIONS: The BELI(E)VE trial aims to establish a novel personalized therapeutic approach for a specific subset of MM patients by combining belantamab mafodotin and venetoclax. This combination targets BCMA and BCL-2, potentially inducing deep and durable remissions. The study will provide critical data on the safety and efficacy of this combination, aiming to improve outcomes in t(11;14)-positive RRMM patients. The translational component, examining circulating tumor cells and mitochondrial activity, may identify mechanisms of resistance and response, guiding future personalized therapeutic strategies.

- Family-centred psycho-oncological counselling for parents with cancer: a longitudinal secondary data analysis of psychological distress, family functioning and parental concerns. [Journal Article]
- CONCLUSIONS: Parents who are confronted with a cancer diagnosis show high levels of psychological distress. Over time, a significant decrease in psychological distress among both groups and family-related outcomes in COSIP users was identified. Interestingly, no significant differences were observed between the groups, suggesting that the type of psycho-oncological support (individual or family-centred) may not differentially affect parental burden.

- Multi-omics profiling reveals sphingolipid metabolism reprogramming of tumor-conditioned MDSCs in cervical cancer. [Journal Article]
- CONCLUSIONS: Our findings uncover sphingolipid metabolic reprogramming as a key mechanism in MDSCs-mediated immune suppression, and propose the Kng1-sphingosine 1-phosphate network as a potential therapeutic target for CC treatment.
