- Imeglimin at clinical doses promotes glucose-stimulated insulin secretion and mitochondrial supercomplex formation. [Journal Article]Endocrinology. 2026 Nov 03. [Online ahead of print]E
- Imeglimin is a novel antidiabetic agent that improves glycemic control by enhancing glucose-stimulated insulin secretion (GSIS). Although previous studies have suggested that imeglimin acts through increased NAD+ production and CD38-dependent calcium signaling, the mechanism responsible for its insulinotropic effect at clinically relevant concentrations remains incompletely understood. In particu…

- Natural fetal testosterone variation is associated with preoptic development and partner preference in male mice. [Journal Article]Endocrinology. 2026 Nov 03. [Online ahead of print]E
- Individual variation in sexually differentiated brain structures may contribute to variation in sex-related behavior, but the developmental origins of such individual differences remain unclear. We first examined several sexually dimorphic nuclei in the preoptic area and bed nucleus of the stria terminalis to identify brain structures associated with individual variation in partner preference in …

- 18F-Fluoro-L-dihydroxyphenylalanine positron emission tomography in the initial staging of medullary thyroid carcinoma. [Journal Article]Minerva Endocrinol (Torino). 2026 Oct 29. [Online ahead of print]ME
- CONCLUSIONS: The role of [18]F-DOPA PET/CT in the initial staging of MTC is selective and context-dependent. While it may provide incremental value in patients with higher tumor burden or suspected lateral nodal disease, its limited sensitivity in the central compartment and in low-volume disease prevents its use as a standalone staging modality. Therefore, [18]F-DOPA PET/CT should be considered a context-driven adjunct rather than a routine tool, to be integrated within a multimodal imaging strategy rather than used as a standalone staging modality.

- Longitudinal effects of gender-affirming hormone therapy on pain, fatigue, and central sensitization-related disorders in individuals with gender dysphoria. [Journal Article]Minerva Endocrinol (Torino). 2026 Oct 29. [Online ahead of print]ME
- CONCLUSIONS: In this study, GAHT was associated with improvements in depressive symptoms but was not associated with significant changes in pain sensitivity or central sensitization measures. Estrogen exposure in AMAB and testosterone therapy in AFAB participants did not lead to measurable changes in CSS-related symptom burden during the follow-up period. However, given the relatively small sample size, these findings should be considered preliminary and require confirmation in larger prospective studies.

- Hepatocyte Pparg knockout and restored Bhmt expression reduce liver steatosis in male mice with diet-induced MASH. [Journal Article]Endocrinology. 2026 Oct 28. [Online ahead of print]E
- Metabolic dysfunction-associated liver disease (MASLD) is characterized by the accumulation of triglycerides in hepatocytes and can progress to metabolic dysfunction-associated steatohepatitis (MASH) with inflammation, hepatocyte ballooning, and fibrosis. MASLD is associated with increased plasma homocysteine levels. Homocysteine is an amino acid generated by the S-adenosylmethionine-dependent tr…

- Hypothalamus-pituitary-adrenocortical responses to critical illness: new insights with clinical implications. [Journal Article]Minerva Endocrinol (Torino). 2026 Oct 23. [Online ahead of print]ME
- Critical illness profoundly affects the hypothalamus-pituitary-adrenocortical (HPA) axis. In the acute phase of sepsis and other critical conditions, systemic cortisol levels rise markedly, yet circulating ACTH is low. Peripheral adaptations - reduced cortisol metabolism, decreased cortisol-binding proteins, and tissue-specific modulation of glucocorticoid receptor signaling - rather than sustain…

- Age and tumor size interact to increase the risk of incomplete response to therapy in differentiated thyroid carcinoma. [Journal Article]Minerva Endocrinol (Torino). 2026 Oct 23. [Online ahead of print]ME
- CONCLUSIONS: In tumors ≤2cm in size, age did not affect the risk of IRT, but when the size is >2cm, advanced age is associated with a higher risk of IRT.

- Food insecurity in the development of metabolic diseases. [Review]
- This Review discusses the current evidence on the relationship between food insecurity, obesity, metabolic syndrome and type 2 diabetes mellitus (T2DM), and identifies potential interventions to mitigate the effect of food insecurity on these conditions. Studies in adults (mainly cross-sectional) have consistently found associations between food insecurity and several cardiometabolic diseases, in…

- Metabolic traits and adaptations of mice lacking Slc12a5 in insulin-secreting β-cells. [Journal Article]Endocrinology. 2026 Oct 21. [Online ahead of print]E
- Pancreatic β-cells integrate metabolic and electrical signals to couple nutrient sensing with insulin secretion, yet the physiological contribution of the neuronal K+Cl- cotransporter-2, Kcc2 (Slc12a5) to β-cell function and whole-body metabolic regulation remains incompletely understood. To define its role in vivo, we generated mice lacking Kcc2 selectively in insulin-secreting β-cells (Kcc2βKO)…

- The β-cells in type 1 diabetes mellitus: findings from human pancreas studies. [Review]
- Type 1 diabetes mellitus (T1DM) is a chronic autoimmune disease characterized by long-term and dynamic interactions between the immune system and pancreatic β-cells, ultimately leading to severe insulin deficiency. A paradigm shift in T1DM research has occurred in the past few years, with research now focusing on the human disease because of access to human pancreatic tissue and pancreas-draining…
