These findings support the potential of prognostic neuroimaging in precision psychiatry and early-risk stratification. However, methodological standardization, external validation, and geographically-diverse samples remain necessary before a full clinical translation.

- CONCLUSIONS: These item-resolved analyses identify widespread but heterogeneous genetic sharing between personality-related phenotypes and psychiatric disorders. The findings provide a pair-specific map of shared loci and prioritized genes, while the Mendelian randomization results should be interpreted cautiously because of residual heterogeneity and potential horizontal pleiotropy. Further validation in diverse populations and functional studies is required.

- CONCLUSIONS: This multinational study provides evidence linking metabolic inflammation, as reflected by RCII, with both depressive symptoms and poor sleep quality. The identification of sleep quality as a partial mediator suggests that sleep disturbance may represent a potential pathway underlying the association between metabolic dysregulation and depression. These findings suggest that RCII may help identify individuals at elevated risk of depressive symptoms, underscoring the importance of integrated strategies targeting metabolic health and sleep in the prevention and management of depressive symptoms.

- CONCLUSIONS: Genetically proxied circulating metabolites showed reproducible associations with MDD, while reverse effects were supported by only two metabolic traits. - CONCLUSIONS: This study provides genetic evidence suggesting medication-specific associations between antidepressant use liability and T2D susceptibility.
