  • Biologic therapy in type 2 CRSwNP: from immunopathology to clinical management. [Review]
    Inflamm Res. 2026 Oct 27; 75(1).He W, Jiang Y, … Wu SIR
  • CONCLUSIONS: Biologic therapies targeting IL-4Rα, IL-5/IL-5R, IgE, and TSLP have demonstrated efficacy in reducing polyp burden, improving symptoms, and decreasing systemic corticosteroid exposure. However, in the absence of robust head-to-head comparisons, optimal patient selection and defining sustainable long-term maintenance strategies remain clinical challenges. Integrating inflammatory profiles, disease burden, and structured follow-up may support individualized treatment and sustained disease control.
  • The networked landscape of cell death in sepsis: from dynamic pathway to node-targeted therapies. [Review]

  • CONCLUSIONS: First, the sequential transformation from early pyroptosis and NETosis (driving hyperinflammation) to mid-stage necroptosis (amplifying tissue damage) to late lymphocyte apoptosis (mediating immunosuppression), with distinct cellular responses in immune cells versus parenchymal tissues. Second, we have distilled complex pathway interactions into three converging decision nodes, RIPK1 ubiquitination status, caspase-8 activity, and membrane pore executors (GSDMD/MLKL/NINJ1), whose coordinated state determines whether cells undergo inflammatory lytic death or silent apoptosis. Third, we describe why single-pathway inhibitors fail in preclinical sepsis studies, and propose a stage-specific therapeutic strategy: inhibit GSDMD early to block pyroptosis; modulate RIPK1 mid-phase to limit RIPK1-dependent inflammatory or necroptotic pathology; and restore lymphoid homeostasis late via caspase-3 inhibition or IL-7 therapy. Finally, we conclude by outlining a translational roadmap prioritising biomarker development, patient stratification, and adaptive trial designs for future research.
  • Immune checkpoint inhibitor-associated myocarditis: clinical and mechanistic insights. [Review]

  • CONCLUSIONS: We highlight the cardiac immune microenvironment as a network of multicellular interactions in which cytotoxic T cells and inflammatory myeloid subsets engage with cardiomyocytes, endothelial cells, and fibroblasts through reinforcing feedback loops. Specifically, interferon-γ (IFN-γ)-driven myeloid activation and chemokine programs (CXCR3-CXCL9/10, CXCL16-CXCR6) coordinate immune cell trafficking and promote tissue injury, while stress-related cell death pathways can trigger innate immune sensing via the cGAS-STING axis. Current management remains centered on early recognition and corticosteroid-based immunosuppression, but emerging mechanistic insights reveal opportunities to improve therapy by targeting immune cell trafficking and inflammatory amplification pathways. Together, these advances support a more integrated cardio-oncology approach to ICIM, in which mechanistic insights inform risk stratification, earlier diagnosis, and the development of more precise immunomodulatory therapies.
