- EasyQuant: An automated imaging-based tool for quantifying neutrophil extracellular traps (NETs). [Journal Article]J Immunol Methods. 2026 Oct 07; :114130. [Online ahead of print]JI
- Neutrophil extracellular traps (NETs) play a crucial role in host defense, yet their dysregulation is associated with inflammation and tissue damage. Consequently, researching NETs in vitro is essential for understanding their role in both health and disease. However, accurate quantification of NETs in vitro remains a challenge due to limitations in manual methods and semi-automated tools, which …

- Efficient detection of human antigen-specific CD4+ T cells with peptide-exchangeable HLA-DP4 tetramers and multiplexed magnetic enrichment. [Journal Article]J Immunol Methods. 2026 Oct 02; :114129. [Online ahead of print]JI
- The tremendous variation in human MHCII alleles is a barrier to using fluorophore-labeled peptide:MHCII tetramers to study antigen-specific CD4+ T cells. This is because matching of the MHCII molecule in the tetramer with each human subject is required. We show that fluorophore-labeled tetramers of peptides complexed to the most common human MHCII molecule-encoded by DPA1*01:03 and DPB1*04:01-det…

- Qualification of ELISA and PRNT methodologies to measure RSV humoral immunity using human clinical samples. [Journal Article]J Immunol Methods. 2026 Sep 27; :114127. [Online ahead of print]JI
- Respiratory syncytial virus (RSV) represents a substantial public health burden worldwide, causing lower respiratory infections primarily in children under 5 and adults over 65. In recent years vaccines and monoclonal antibodies (mAbs) have demonstrated promising prophylaxis against RSV in vulnerable populations, benefitting individuals, families, and healthcare systems. However, this protection …

- How many gamma delta T cells are there? Calling strategy, quality control and receptor-locus specificity in 3-prime single-nucleus RNA sequencing of human small intestine. [Journal Article]J Immunol Methods. 2026 Sep 27; 551:114126. [Online ahead of print]JI
- Public single-nucleus transcriptomes are increasingly reused to count gamma delta T cells in the human small intestine, but how far that count depends on analytical choices has not been quantified. Using a public 3-prime single-nucleus dataset of 33,600 nuclei from six small-intestinal biopsies, we benchmarked three calling strategies across three quality-control levels, tested six module-score c…

- Mutations in the Iduronate 2-sulfatase (IDS) gene cause Hunter syndrome (also known as mucopolysaccharidosis type II, or MPS II), a rare genetic condition that causes glycosaminoglycans (GAGs) to accumulate. Although enzyme replacement therapy (ERT) is the usual course of treatment, it can cause immunological reactions, such as the generation of IgE antibodies specific to the drug, which can resu…

- Isotype-specific optimization of antigen-specific ELISA reveals differential effects of blocking reagents. [Journal Article]J Immunol Methods. 2026 Sep 17; 551:114125. [Online ahead of print]JI
- Accurate detection of antigen-specific antibodies by ELISA requires minimizing nonspecific background, yet blocking conditions are rarely optimized for antibody isotype. Here, we systematically evaluated the impact of blocking reagents on isotype-specific detection of antibodies against influenza A/PR8 hemagglutinin (HA) in mice using an antigen-specific indirect ELISA with isotype-specific detec…

- Lipemia and serum index-related interference in Luminex-based anti-HLA antibody detection. [Journal Article]J Immunol Methods. 2026 Sep 03; 551:114122. [Online ahead of print]JI
- CONCLUSIONS: Lipemia represents an underrecognized source of interference in Luminex-based HLA antibody testing. Automated HIL indices alone did not capture all analytically compromised samples in this cohort. Systematic monitoring of internal control bead values may improve detection of silent and index-independent interferences and enhance analytical accuracy.

- A novel multiplex serological immunoassay for health monitoring in rodents. [Journal Article]J Immunol Methods. 2026 Sep 02; 551:114123. [Online ahead of print]JI
- Health monitoring of laboratory rodents is essential for ensuring animal welfare, meeting regulatory requirements for zoonosis control, and maintaining the integrity and reproducibility of scientific studies. Routine serological screening, conducted in accordance with FELASA guidelines, plays a critical role in the detection of infectious agents. While conventional serological assays typically ta…

- Development and diagnostic evaluation of indirect ELISA for detecting antibodies against the rabies virus glycoprotein in human cerebrospinal fluid. [Journal Article]J Immunol Methods. 2026 Sep 01; 551:114113. [Online ahead of print]JI
- The accurate detection of intrathecal antibodies against the rabies virus (RABV) is critical for antemortem diagnosis and disease monitoring, particularly when the virus-neutralizing activity is low or delayed. In this study, indirect ELISA assays were developed to detect IgM, IgG, and polyvalent (IgA/IgM/IgG) antibodies against the RABV glycoprotein (RABV G) in cerebrospinal fluid. The RABV G an…

- Mycobacterium tuberculosis (M. tb), the causative agent of tuberculosis (TB), remains the leading cause of death from a single infectious agent. The only approved vaccine for TB, Bacille Calmette-Guerin (BCG), provides variable and incomplete protection from disease, underscoring the need to improve vaccine development. Preclinical murine models are widely used to triage TB vaccine candidates, bu…

- Thymic epithelial cells (TECs) play a vital role in the generation of immunocompetent and immunotolerant T cells. The understanding of TEC biology in model animals such as mice has advanced thanks to the cellular and molecular analyses of purified TEC populations, including cortical TECs (cTECs) and medullary TECs (mTECs). On the other hand, studies of human TECs have fallen behind because of the…

- Although most available therapeutic antibodies are immunoglobulin (Ig) G, their limited access at mucosal sites has prompted increasing interest in alternative isotypes, such as IgM. Therapeutic IgM antibodies have primarily been obtained from patient sera; however, methods for generating IgM antibodies that specifically target antigenic regions remain poorly established. In the present study, we…

- Dynamic effects of short-term storage temperature and duration on the transcriptome and functional pathways of umbilical cord blood-derived NK cells. [Journal Article]J Immunol Methods. 2026 Sep; 550:114110.JI
- CONCLUSIONS: CBMC-derived NK cells can tolerate up to 24 h of ex vivo storage while maintaining transcriptome and core functional stability. However, extending the ex vivo storage period to 72 h leads to significant reprogramming in the expression of immune and metabolism-related genes. Notably, the degree of metabolic suppression under refrigerated conditions at 4 °C was less pronounced than that at 25 °C room temperature storage. This study recommends that the ex vivo storage time of UCB should ideally be limited to within 24 h, with room temperature storage being a viable option. If prolonged storage is necessary, refrigeration at 4 °C is advised.

- Instrument-specific reference intervals for coagulation factor XII and antithrombin III: A regional study in Jiangsu, China. [Journal Article]J Immunol Methods. 2026 Aug 27; 551:114112. [Online ahead of print]JI
- CONCLUSIONS: This study established and verified regional RIs for FXII and ATIII in healthy adults from Jiangsu, China. Although analyzer-related differences between the CS-5100 and CS-6500 platforms were modest, the findings highlight the importance of local RI verification when applying manufacturer-derived intervals or changing analytical platforms. These locally derived RIs may improve the interpretation of FXII and ATIII results and reduce unnecessary follow-up testing caused by inappropriate generic intervals.
