  • The CLC family of chloride channels and transporters plays crucial roles in endosomal/lysosomal function, with dysfunction contributing to protein aggregation and dementia development. CLCN6 mutations are recognized in early-onset neurodegenerative disorders, but reports from India remain limited. We report the first case from India of frontotemporal dementia associated with a novel CLCN6 mutatio…
  • Can neurodevelopmental disorders be part of brain-lung-thyroid syndrome? A case report. [Journal Article]

  • Mutations in the NKX2-1 gene cause a syndromic disorder affecting the brain, thyroid, and lungs. There are few case reports in the literature discussing the association between NKX2-1 gene mutation and neurodevelopmental disorders. In this case report, a 12-year-old girl with an NKX2-1 gene mutation, articulation disorder, and specific learning disorder is presented. This report discusses the pot…
  • A de novo 1.62 Mb deletion at 2q34 with nonpenetrant neurodevelopmental phenotype at 12 months. [Journal Article]

  • CONCLUSIONS: This report describes a de novo 2q34 deletion involving ERBB4, a gene strongly linked to intellectual disability and epilepsy. However, the absence of any neurodevelopmental abnormalities at 12 months suggests that this deletion may exhibit incomplete penetrance and variable expressivity. This case highlights the complexity of genotype-phenotype correlation for ERBB4 haploinsufficiency and underscores the value of long-term follow-up.
  • A rare missense variant in Bruton's tyrosine kinase is associated with bipolar disorder accompanied by psychosis. [Journal Article]

  • CONCLUSIONS: Most reported BTK variants are known to cause agammaglobulinemia. A de-novo variant in BTK p.(Arg255Gln) was previously associated with schizophrenia in one female patient. Multigenic deletions, including BTK , can cause deafness-dystonia-optic neuropathy syndrome, sometimes accompanied by psychosis. Taken together, these studies indicate that BTK may have a role in some mental disorders that needs to be investigated further. Use of animal models may elucidate any BTK function in the brain.
