Inadequate concentrations of the HIV protease inhibitor saquinavir jeopardize the individual therapy success or produce side effects despite treatment according to the current guidelines. We performed a population pharmacokinetic analysis with NONMEM and indentified that steady-state saquinavir pharmacokinetics in 136 HIV-1 infected adults were modulated by a decrease in saquinavir clearance following co-administration of CYP3A inhibitors ritonavir and atazanavir. In contrast, age, sex, weight, pregnancy or the pharmaceutical formulation exerted only minor non-significant effects.
