- Pre-existing cross-subtype serological responses against H5Nx in health care workers. [Journal Article]J Virol. 2026 Oct 07; :e0131026. [Online ahead of print]JV
- A rise in cases of highly pathogenic avian influenza A (H5N1) clade 2.3.4.4b in humans has been reported since 2023. Although influenza seasonal vaccination (H1N1/H3N2) was shown to be effective against cases of reinfection with endemic influenza A viruses (IAVs), its capacity to elicit cross-reactive antibodies against (H5N1) remains unclear. Here, we measured vaccine-elicited antibody responses…

- Exploring the immunological effects of antibody-drug conjugates. [Review]
- Antibody-drug conjugates (ADCs) have profoundly reshaped the therapeutic landscape across multiple malignancies, establishing a new paradigm of targeted cytotoxic delivery. Emerging evidence indicates that the activity of ADCs extends beyond direct cytotoxicity to encompass a coordinated spectrum of immunomodulatory effects, arising from both antibody-mediated functions and payload-induced cellul…

- Why antibody isotype matters for immunotherapy. [Review]
- Monoclonal antibodies (mAbs), engineered to recognize disease-associated antigens, are central to cancer immunotherapy due to their ability to block or eliminate malignant cells by engaging immune effector mechanisms. Yet many tumors evade or suppress these responses, limiting durable clinical benefit and highlighting the need to optimize antibody-based strategies. A critical determinant of mAb a…

- A fully human afucosylated anti-GARP antibody PBA-0111 drives anti-tumor immunity through combined ADCC induction and TGF-β1 pathway blockade. [Journal Article]
- CONCLUSIONS: These results underscore the therapeutic potential of PBA-0111 as a dual-functioning immunotherapy and a promising candidate for the treatment of GARP-positive cancers. This integrated mechanism may amplify anti-tumor immunity, improve synergy with checkpoint inhibitors, and provide more durable reprogramming of the tumor microenvironment than either approach alone.

- Vpu protects HIV-infected cells from antibody-dependent cellular phagocytosis by counteracting tetherin. [Journal Article]Cell Rep. 2026 Sep 30; 45(10):118080. [Online ahead of print]CR
- Tetherin is an interferon-inducible transmembrane protein that inhibits the detachment of enveloped viruses from infected cells. HIV-1 Vpu counteracts this restriction by removing tetherin from sites of virus release. Accordingly, Vpu mutations that disrupt tetherin antagonism increase Env accumulation on the surface of HIV-infected cells. We found that Vpu mutations that impair tetherin antagoni…

- Natural Killer Cells in Kidney Transplantation: Dual Role in Allograft Injury and Immune Adaptation. [Review]Arch Immunol Ther Exp (Warsz). 2026 Jan 01; 74(1).AI
- Kidney transplantation is the optimal treatment for end-stage renal disease, yet long-term graft survival remains limited by allograft rejection. While T and B cells and donor-specific antibodies (DSA) have been the main focus, accumulating evidence indicates that natural killer (NK) cells play a significant dual role in both graft injury and immune adaptation, including in DSA-negative rejection…

- Selinexor potentiates NK cell activation against multiple myeloma. [Journal Article]Clin Exp Immunol. 2026 Sep 26. [Online ahead of print]CE
- CONCLUSIONS: These data reveal that selinexor selectively disrupts the NKG2A:HLA-E immune checkpoint axis in MM and enhances NK cell activation and ADCC against MM cells. These findings provide a mechanistic rationale for investigating combinations of selinexor with approved therapeutic antibodies in MM and suggest that treatment timing may be an important consideration in the design of combination regimens.

- TLR8 Stimulation Leads to Microenvironment Activation and Enhanced Antibody-Dependent Cellular Responses in CLL. [Journal Article]
- Toll-like receptors (TLRs) are crucial for eliciting immune responses against pathogens such as bacteria and viruses. TLR activation has also been studied for its potential to drive antitumor activity in immune cells. TLR8 is an intracellular receptor that recognizes single-stranded RNA and promotes antiviral responses, including the release of cytokines such as TNFα and type I IFN. Previously we…

- IMGT[®], the international information system[®] (IMGT), was created in 1989 by Marie-Paule Lefranc (Université de Montpellier and CNRS) in Montpellier, France, to deal with and to manage the huge diversity of immunoglobulins (IG) or antibodies and T-cell receptors (TR), which are the antigen receptors (AR) of the adaptive immune response (AIR) of jawed vertebrates. The founding of IMGT[®] marked…

- Phenotypic Plasticity of Tumor-Associated Neutrophils in the Solid Tumor Microenvironment and Emerging Interventions. [Journal Article]Curr Med Chem. 2026 Sep 23. [Online ahead of print]CM
- Tumor-associated neutrophils (TANs) are a highly dynamic and key component of the tumor microenvironment (TME) in solid tumors and are characterized by an exceptional degree of phenotypic plasticity and functional heterogeneity. TANs play an important role in the control of tumor evolution, metastasis, immune suppression, and therapeutic resistance. The heterogeneity of TANs is reflected in their…

- Tumor Acidosis Boosts Antibody-Driven Cytotoxic Potential of Neutrophils. [Journal Article]
- The acidic tumor microenvironment (pH 6.3-7.1) influences immune cell function, yet its impact on antibody-mediated killing remains poorly understood. While natural killer (NK) cells lose their spontaneous tumor-killing ability in acidic conditions, the effect on IgG-mediated killing is unexplored. Neutrophils, key effector cells in IgA immunotherapy, respond to pH changes by delayed apoptosis an…

- Structural basis of diverse antibody recognition of conserved coronavirus spike S2 epitopes that contribute to protective immunity. [Journal Article]
- Conserved epitopes within the coronavirus spike S2 domain elicit broadly reactive antibodies, yet many characterized responses show limited neutralizing and variable protective activity, leaving their contribution to antiviral immunity unclear. Building on our previous mapping of evolutionarily conserved spike "coldspots", we isolated human monoclonal antibodies targeting four conserved epitopes …

- Antibody-Dependent Cellular Phagocytosis and Cytotoxicity in Patients With LGI1 and CASPR2 Encephalitis. [Journal Article]
- CONCLUSIONS: ADCC and ADCP, but not CDC, are common effector functions of LGI1/CASPR2-IgG, and their presence correlates with long-term poor outcome, suggesting that they might represent a useful prognostic biomarker and suggest additional pathogenic mechanisms. We provide the proof- of principle for a framework that could be applied to other autoantibody-mediated disorders.

- Sinonasal POU2AF3 (COLCA2)-rearranged carcinomas: a study of 11 aggressive tumors with findings supporting classification as solid adenoid cystic carcinoma. [Journal Article]
- POU class 2 homeobox associated factor (POU2AF3) is a gene that has been associated with increased risk of colon cancer. Recently, colleagues have reported recurrent EWSR1/FUS::POU2AF3 fusions in undifferentiated spindle cell/round cell sarcomas with a predilection for the sinonasal tract. We report 11 sinonasal POU2AF3-rearranged carcinomas that are distinct from the EWSR1/FUS::POU2AF3 sarcomas.…
