  • Cancer plasticity and histological transformation as adaptive resistance mechanisms in precision oncology. [Review]
Med. 2026 Oct 13; :101326. [Online ahead of print]Kuburich NA, Gouda MA, … Subbiah VMED
  • The advent of precision oncology has brought substantial advancements in cancer therapy, yet it has been accompanied by equally complex resistance mechanisms. Among these mechanisms, histological transformations can compromise the effectiveness of targeted therapies, resulting in a loss of clinical benefit following initial success. Understanding these evolutionary processes is crucial for develo…
  • Hitting KRAS G12D without the gut punch: Can immunotherapy make it last? [Journal Article]
    Cancer Cell. 2026 Oct 12; 44(10):1921-1923.Kim DW, Garrido-Laguna ICC
  • In this issue of Cancer Cell, Shi et al. report encouraging early-phase results combining HRS-4642, an allele-specific KRAS G12D inhibitor, with the PD-L1 inhibitor adebrelimab in previously treated metastatic pancreatic cancer. The intravenous combination was safe and well tolerated, notably sparing patients the gastrointestinal toxicity common to oral KRAS inhibitors, and achieved a promising 4…
  • A modular framework for antibody-drug conjugate biomarkers. [Review]
Nat Rev Clin Oncol. 2026 Oct 09. [Online ahead of print]Liu J, Ruan DY, … Pistilli BNR
  • The availability of approved antibody-drug conjugates (ADCs) has rapidly expanded across oncology indications, yet biomarker development remains dominated by semi-quantitative immunohistochemistry. Robust biomarkers are needed to improve patient selection, guide therapeutic sequencing and identify resistance mechanisms that inform rational combinations. ADC activity depends on target antigen expr…
  • da Vinci Single-Port Transoral Robotic Surgery in European Practice: Early Outcomes and Comparison With Transoral Laser Microsurgery. [Journal Article]
Clin Otolaryngol. 2026 Oct 08. [Online ahead of print]Ryan ME, Zammit M, … Fleming JCCO
  • CONCLUSIONS: SP-TORS implementation in a high-volume UK centre was feasible and safe during early adoption, with low complication rates. Peri-operative and early functional outcomes were broadly comparable with TLM. Although specimen ink-positive margins were less frequent following SP-TORS, interpretation should be cautious given the historical comparator and associated differences in departmental workflow. These findings support safe integration of SP-TORS into transoral surgical practice and justify prospective evaluation.
  • Updates in CAR-T and TCR-T therapy for solid tumors from the 2026 ASCO Annual Meeting. [Letter]
    J Hematol Oncol. 2026 Oct 07; 19(1).Mirallas O, Dumbrava EEJH
  • Adoptive cellular therapies (ACT) have reshaped the treatment landscape of hematologic malignancies and are increasingly being explored in solid tumors. Key approaches include chimeric antigen receptor T-cell (CAR-T) therapy, which targets tumor-associated surface antigens in an HLA-independent manner, and T-cell receptor-engineered T-cell (TCR-T) therapy, which enables recognition of intracellul…
  • Non-Conventional Histologies of Prostate Cancer: From Morphology to Biology-Driven Precision Oncology. [Journal Article]
    Oncologist. 2026 Oct 05. [Online ahead of print]Flauto F, Neola G, … Banna GLO
  • Non-conventional prostate cancer histologies represent a small but clinically significant subset characterised by aggressive behaviour, atypical biomarker profiles, and reduced dependence on androgen receptor signalling. Evidence indicates these histologies reflect dynamic biological states driven by genomic instability, lineage plasticity, and therapeutic selection rather than static morphologic…
  • Redefining breast cancer treatment by precision oncology: the clinical impact of sacituzumab govitecan. [Review]
Expert Opin Biol Ther. 2026 Oct 07; :1-10. [Online ahead of print]Di Lauro V, Sacco PC, … Rizzo AEO
  • Trophoblast cell-surface antigen-2 (Trop-2) is overexpressed in most breast cancers, including about 90% of triple-negative breast cancers (TNBC) and more than half of hormone receptor-positive/HER2-negative (HR+/HER2-) tumors, with limited expression in normal tissues, making it an attractive therapeutic target. Sacituzumab govitecan (SG), the first Trop-2-directed antibody-drug conjugate (ADC) …
