- Zerumbone alleviates TRESK- and TRPV1-mediated ion currents and reduces excitability of human iPSC-derived nociceptor-like neurons. [Journal Article]Mol Pain. 2026; 22:17448069261492873.MP
- Secondary plant metabolites are widely used in traditional and naturopathic medicine to treat pain, infection, and inflammation. The sesquiterpenoid zerumbone, derived from the edible ginger Zingiber zerumbet, has been shown to exert analgesic, antioxidant, antitumoral, and antimicrobial effects in preclinical models, acting through multiple receptor systems including cannabinoid (CB) receptors, …

- Modulation of nociceptive nerve activity as a novel therapeutic strategy in acute myeloid leukemia. [Journal Article]Fundam Res. 2026 Sep; 6(5):3507-3518.FR
- Emerging evidence has highlighted a critical role of the neuro-microenvironment in regulating hematopoiesis and hematologic malignancies. However, how malignant cells remodel the neural compartment and how these neural changes in turn influence disease progression remain poorly understood. In this study, we mapped the spatial distribution of sympathetic (TH+) and nociceptive (CGRP+) nerve fibers …

- Insulin Enhances Nitroglycerin-Induced Hyperalgesia Through Insulin-Responsive Receptor Signalling Involving the Trigeminovascular System and Meningeal Mast Cells. [Journal Article]Eur J Neurosci. 2026 Oct; 64(7):e70705.EJ
- Migraine headache prevalence is lower in patients with diabetes, but the underlying mechanisms remains unclear. This study explored the influences of hyperglycemia, insulin and insulin receptor (IR)/insulin-like growth factor 1 receptor (IGF-1R)-related signalling on migraine-related mechanisms in in vivo and ex vivo rat models. In in vivo experiments, migraine-relevant responses were induced by …

- Effects of isolated and combined supplementation of caffeine and capsaicin on a self-paced 10-km running performance. [Journal Article]
- CONCLUSIONS: Combined CAF and CAP reduced 10-km completion time and increased post-exercise blood lactate and perceived vigor versus placebo. Session-RPE and heart rate did not differ among conditions; RPE differed only between CAF and CAP. CAF-containing conditions also produced more frequent transient adverse effects.

- Clinical Outcomes Associated with the use of the 8% Capsaicin Patch in Localized Neuropathic Pain: A Quasi-Experimental Study. [Journal Article]Enferm Clin (Engl Ed). 2026 Oct 05; :502533. [Online ahead of print]EC
- CONCLUSIONS: The use of the 8% capsaicin patch was associated with improvements in pain and related outcomes and showed a favourable safety profile. Nurses play an important role in the application, patient education, and follow-up of this intervention. Specific training in this intervention could improve care quality and patient outcomes. Further studies should assess its long-term efficacy and combination with systemic therapies.

- Bioinformatics-based identification of capsaicin target genes associated with trastuzumab resistance in HER2-positive breast cancer: in silico and qRT-PCR validation studies. [Journal Article]In Silico Pharmacol. 2026; 14(3):253.SP
- Trastuzumab (TRZ) is a primary treatment for HER2-positive (HER2+) breast cancer, but its effectiveness is often limited by drug resistance linked to the PI3K signaling pathway. Capsaicin (CAP), a natural compound from chili peppers, has been shown to block the PI3K/AKT pathway. This study aimed to computationally identify specific gene targets of CAP and clarify its molecular mechanism for overc…

- Discovery and Characterization of Neuroactive Secondary Metabolites from Acacia sieberiana with Potential Analgesic Properties. [Journal Article]
- Acacia sieberiana (syn. Vachellia sieberiana), widely used in African traditional medicine for pain, was investigated using an ethnopharmacology-guided workflow integrating LC-MS/MS-based metabolomics and molecular networking to prioritize bioactive secondary metabolites. Fractionation of the ethanolic stem bark extract enabled targeted isolation and identification of piperine (1) and moupinamide…

- NRF2 activation mediates the protective effects of capsaicin against MGO/STZ-induced chondrocyte apoptosis and diabetic articular cartilage injury. [Journal Article]Toxicol Appl Pharmacol. 2026 Oct 01; 517:118068. [Online ahead of print]TA
- Diabetes mellitus is an independent risk factor for osteoarthritis (OA), and the molecular mechanisms underlying diabetic cartilage damage remain unclear. This study investigated the role of oxidative stress in diabetes-induced chondrocyte injury, especially chondrocyte apoptosis, explored the protective role of the NRF2 antioxidant system, and evaluated the therapeutic potential of capsaicin for…

- Effects of acute Capsaicin analogue intake on athletic performance: a three-level meta-analysis. [Meta-Analysis]
- CONCLUSIONS: Acute capsaicin analogue intake may produce small but significant improvements in athletic performance, particularly in power and muscular endurance. However, due to heterogeneity, publication bias, and very low certainty of evidence, these findings should be interpreted cautiously.

- Assessing the value of systematic screening and early management of neuropathic pain after total knee arthroplasty. A single-center retrospective study. [Journal Article]Orthop Traumatol Surg Res. 2026 Sep 30; :104914. [Online ahead of print]OT
- CONCLUSIONS: The observed NP rate and CPOP risk-factor profile were consistent with the literature. Nearly two-thirds of analyzed patients met the predefined recovery criteria following systematic screening and multidisciplinary management. However, the absence of a control group precludes causal attribution of these outcomes to the pathway. Prospective controlled studies are required to determine its effect on recovery and symptom control.

- Self-assembled nanoparticle system synchronizing rapid analgesia and sustained anti-inflammatory therapy for inflammatory pain. [Journal Article]Int J Pharm. 2026 Sep 28; 705:127478. [Online ahead of print]IJ
- Inflammatory pain severely impacts patient well-being. Current treatments focus heavily on resolving inflammation yet often fail to offer rapid pain relief, leading to poor clinical compliance. To bridge this gap, we designed self-assembled nanoparticles that pair instant pain relief with long-term control of inflammation. The core consists of co-assembled lidocaine (LDC), capsaicin (CAP), and me…

- Ketamine and Sodium Selenite Promote Transient Receptor Potential Vanilloid 1-Associated Calcium Signaling and Apoptosis in SH-SY5Y Neuroblastoma Cells. [Journal Article]
- Ketamine has pharmacological actions beyond N-methyl-D-aspartate receptor antagonism, whereas sodium selenite modulates redox-dependent signaling. Transient receptor potential vanilloid 1 (TRPV1) channels regulate Ca[2+] influx and may contribute to oxidative stress-associated apoptosis. We investigated whether ketamine and sodium selenite, alone and combined, promote apoptosis through TRPV1-asso…
