- Impact of surface structuring on antibiotic elution from bone cement in two-stage revision for periprosthetic joint infection. [Journal Article]
- CONCLUSIONS: Surface modification of PMMA cement influenced antibiotic elution in this in vitro experimental model, with increased surface depth resulting in enhanced vancomycin release. Gentamicin elution was less affected by surface geometry and showed significant improvement only with the greatest surface enlargement. These findings indicate that structured cement surfaces may influence local antibiotic release characteristics under controlled laboratory conditions. However, the clinical relevance of these surface modifications remains to be determined, as this study was limited to cement blocks incubated in phosphate-buffered saline and did not include a spacer, infection, or in vivo model. Further experimental and in vivo investigations are required to assess whether surface structuring can improve antibiotic delivery in clinically relevant settings.

- Lateral line - evoked potentials (LLEPs): an effective tool for investigating ototoxicity. [Journal Article]Toxicol Appl Pharmacol. 2026 Nov; 516:118041.TA
- Investigations of ototoxicity using the zebrafish lateral line system have focused on the morphology of neuromast sensory hair cells using vital dyes. As a complement to this approach, we demonstrate the utility of lateral line - evoked potentials (LLEPs). The LLEPs were acquired with a modified auditory evoked potentials physiological rig, using a dipole stimulus that was presented at a rate of …

- Gentamicin Targeting Human Hemoglobin Induces Methemoglobin Formation and Decreases Oxygen Affinity: A Molecular Mechanism of Hematologic Toxicity. [Journal Article]Int J Mol Sci. 2026 Aug 29; 27(17).IJ
- The nephrotoxicity and ototoxicity of Gentamicin have been extensively investigated; however, whether they induce hematotoxicity remains unclear. This study aimed to explore the binding of Gentamicin to adult hemoglobin (HbA) and its toxicological implications. The effect of Gentamicin on HbA oxidation was assessed by quantifying methemoglobin (MetHb) formation via a four-wavelength spectrophotom…

- Dual Release of Antibiotics with Additive Effect from Cleavable Hydrogels Based on Amino/Thiol-Yne Click Chemistry. [Journal Article]
- Dual-antibiotic loaded poly(ethylene glycol) (PEG)-based hydrogels were developed as antimicrobial wound dressings combining covalent and physical antibiotic incorporation. Hydrogels were prepared through amino-yne click conjugation of gentamicin sulfate (Gs) and thiol-yne cross-linking, followed by ciprofloxacin (CPX) loading via swelling-diffusion. The materials exhibited tunable gelation time,…

- Enhanced Antimicrobial and Immunomodulatory Effects of Calcium Hydroxide Combined with NSAIDs Against Enterococcus faecalis: An In Vitro Study. [Journal Article]Eur Endod J. 2026 Jul 22; 11(7):319-329.EE
- CONCLUSIONS: Combining Ca(OH)2 with diclofenac sodium or Keto enhances the antimicrobial and immunomodulatory efficacy of Ca(OH)2 against E. faecalis biofilms. Ca(OH)2 + diclofenac reduced PBMCs viability, suggesting a strong immunosuppressive effect that may be beneficial in dampening excessive inflammation but also raises concerns regarding cytocompatibility and dose optimization. Ca(OH)2 + keto demonstrated a more favourable balance between antimicrobial activity and biocompatibility, supporting its potential as a non-antibiotic adjunct in intracanal medication protocols.

- One Model to Dose Them All: A Population Pharmacokinetic Model for Multiple Aminoglycosides Across the Human Lifespan. [Journal Article]
- Aminoglycosides require therapeutic drug monitoring due to substantial pharmacokinetic variability across individuals. While population pharmacokinetic (popPK) models have been developed for individual drugs across age groups, and cross-drug maturation models have been explored, it is unclear whether pooling data across aminoglycosides improves predictive performance. We developed and evaluated a…

- Deciphering gentamicin-induced nephrotoxicity and hepatotoxicity: Pathophysiological insights and emerging protective approaches. [Review]Food Chem Toxicol. 2026 Nov; 217:116350.FC
- Gentamicin (GEN) is a critical aminoglycoside antibiotic for severe gram-negative infections, yet its clinical utility is limited by dose-dependent nephrotoxicity (occurring in 10-25% of therapeutic courses) and hepatotoxicity. Nephrotoxicity arises from GEN accumulation in proximal tubular cells, triggering reactive oxygen species overproduction, lipid peroxidation, mitochondrial dysfunction, an…

- Gentamicin, a widely used aminoglycoside antibiotic, causes severe nephrotoxicity and ototoxicity through ferroptosis-mediated mechanisms involving glutathione peroxidase 4 (GPX4), the cystine/glutamate antiporter SLC7A11, NADPH oxidase 4 (NOX4), and Prestin (SLC26A5). Ferrostatin-1, a selective ferroptosis inhibitor, demonstrates preclinical cytoprotective efficacy but lacks antimicrobial activi…
