- Promethazine use and QT prolongation risk during treatment with a shortened all-oral regimen for MDR/RR-TB. [Journal Article]JAC Antimicrob Resist. 2026 Oct; 8(5):dlag215.JA
- CONCLUSIONS: Promethazine use was associated with increased risk of clinically relevant QTc prolongation within a QTc-burdened all-oral MDR/RR-TB regimen. These findings provide preliminary evidence that clinicians may consider when selecting anti-emetics as part of MDR/RR-TB therapy with a shortened all-oral regimen and highlight the importance of evaluating adjunctive medications in DR-TB care.

- Zuclopenthixol acetate for acute schizophrenia and similar major mental illnesses. [Systematic Review]
- CONCLUSIONS: Based on the evidence in this review, there may be no difference between zuclopenthixol acetate and standard medications in the management of acute psychiatric symptoms, tranquillisation and adverse events. Zuclopenthixol acetate was probably associated with a greater degree of sedation at 24 and 48 hours of administration. The use of zuclopenthixol acetate may be associated with a lower requirement for adjunctive benzodiazepines, but this is uncertain. Doses of zuclopenthixol acetate of 25 mg to 50 mg per injection may be as effective as doses of 50 mg to 100 mg per injection (very low-certainty evidence). Recommendations favouring zuclopenthixol acetate over standard medications for the management of acute behavioural disturbance should be interpreted with caution, given the methodological limitations of most included studies.

- Comparison of the Utility of Amplitude-Spectral and Coherence Features of Psychotropic Drugs' Action on ECoG Signal for Pharmaco-EEG Based Drug Screening in Rats. [Journal Article]
- A naive Bayesian classifier (NBC) combined with principal component analysis (PCA) effectively differentiates the dose-dependent effects of certain groups of psychoactive drugs based on their impact on the amplitude-spectral characteristics of electrocorticograms (ECoG) in rats. This approach has been shown to be useful for pharmacological screening of agents with unknown or poorly understood act…

- Method Development and Validation of Promethazine HCl by Using RP-HPLC and Its Forced Degradation Studies. [Journal Article]Assay Drug Dev Technol. 2026 Sep 22; :1540658X261486549. [Online ahead of print]AD
- A simple, rapid, sensitive, and stability-indicating reverse-phase high-performance liquid chromatography (RP-HPLC) method was developed and validated for the quantitative determination of promethazine hydrochloride (HCl) in pharmaceutical formulations. Chromatographic separation was achieved using a SunFire® C18 column (4.6 × 250 mm, 5 µm) with a mobile phase consisting of acetonitrile and 0.1% …

- Efflux pump inhibition disrupts biofilms and enhances azole susceptibility in Candidozyma auris. [Journal Article]Biofouling. 2026 Oct; 42(9):1039-1051.B
- Candidozyma auris (formerly Candida auris) is a global emerging fungal pathogen of significant global clinical and epidemiological concern. It is frequently associated with hospital outbreaks, largely due to its multidrug resistance, ability to persist on medical devices, and high potential for nosocomial transmission. This study investigated efflux pump inhibition as a potential strategy for con…

- Antiemetic-Associated QT Prolongation: A Clinical Review and Risk-Stratified Prescribing Framework for Long QT Syndrome. [Journal Article]Eur Heart J Cardiovasc Pharmacother. 2026 Sep 16. [Online ahead of print]EH
- CONCLUSIONS: Antiemetic-associated arrhythmia is largely preventable. Structured risk assessment, electrolyte correction, ECG monitoring, and a stepwise selection strategy favouring agents with minimal QT liability allow clinicians to manage nausea and vomiting safely, even in high-risk populations.

- Regulation of Template Ionization and Template-Monomer Interactions for Enhanced Molecular Imprinting and Selective Detection of Promethazine. [Journal Article]Polymers (Basel). 2026 Sep 02; 18(17).P
- A molecularly imprinted polymer (MIP)-based quartz crystal microbalance (QCM) sensor was developed for the selective detection of promethazine (PMZ) through deliberate regulation of template-monomer interactions during the molecular imprinting process. Various functional monomer systems were systematically evaluated to optimize intermolecular recognition, among which the combination of 4-vinylpyr…

- A hERG blocker facilitates K+ channel current by promoting pore opening while blocking. [Journal Article]
- Many drugs that block voltage-gated K+ channels encoded by the human ether-à-go-go-related gene (hERG) can cause long QT syndrome and life-threatening cardiac arrhythmias, yet the molecular mechanisms that determine this risk remain unclear. A process that may counteract arrhythmogenic hERG block, termed facilitation, is common to many clinically approved hERG blockers, including nifekalant, amio…

- Verapamil-sensitive fascicular ventricular tachycardia precipitated by promethazine overdose. [Case Reports]BMJ Case Rep. 2026 Aug 24; 19(8).BC
- Verapamil-sensitive fascicular tachycardia is a type of ventricular tachycardia (VT) involving the specialised cardiac conducting tissue. Drug overdose is an important cause to consider when assessing patients with a new presentation of a broad complex tachycardia. Multiple antihistamines have been implicated as pro-arrhythmic, but these tend to precipitate Torsades de Pointes via drug-induced QT…

- Antiemetic Combination Use During the First Trimester and Adverse Circulatory Outcomes in Newborn Infants: A Data-Mining Analysis. [Journal Article]
- CONCLUSIONS: By scanning more than 300 infant circulatory-related outcomes, we identified unspecified hypotension as a potential safety signal among newborns whose mothers were exposed to promethazine-ondansetron combination rather than metoclopramide-ondansetron combination during the first trimester. This finding warrants further investigation and illustrates the utility of tree-based scan statistics as a hypothesis-generating tool for pregnancy medication safety research.

- .Degradation of active drugs in medicinal vials exposed to temperature extremes encountered during military operations: the Military Medication Temperature Effect Study. [Journal Article]BMJ Mil Health. 2026 Aug 11. [Online ahead of print]BM
- CONCLUSIONS: Most of the medications tested retained acceptable physicochemical characteristics over 28 days under conditions simulating field exposure. These findings provide preliminary support that some medications may remain suitable for use when manufacturer-recommended temperatures cannot be maintained, and resupply is unlikely. However, the single sample (n=1) study design limits confidence in the findings. Further replicated studies are required before these results should inform medicines management policy.

- Analysis of eight OTC drug components in postmortem dried blood spot samples by liquid chromatography-tandem mass spectrometry. [Case Reports]Leg Med (Tokyo). 2026 Sep; 85:102954.LM
- Recently, abuse and fatal overdoses of over-the-counter (OTC) medications have become serious social and health issues worldwide. However, many drug components in OTC medications that are abused have different chemical structures from those assessed by commercially available rapid drug test kits. In this research, we focused on using dried blood spot (DBS) samples to develop a simple and accurate…
