- Modified clomiphene citrate protocol versus gonadotropin-releasing hormone antagonist protocol in in vitro fertilization: a propensity score-matched retrospective study on cost and efficacy. [Journal Article]Front Endocrinol (Lausanne). 2026; 17:1888569.FE
- CONCLUSIONS: The modified CC protocol was associated with ovarian stimulation effects and clinical outcomes comparable to those of the GnRH-ant protocol. Furthermore, it represented a more cost-effective ovarian stimulation strategy, significantly reducing treatment costs while maintaining a clinical pregnancy rate comparable to that of the GnRH-ant protocol.

- Micro-TESE in Non-Obstructive Azoospermia: Phenotype-Guided Hormonal Optimization and Testosterone Response-Prognostic Biomarker or Therapeutic Target? [Review]
- Non-obstructive azoospermia (NOA) is the most severe phenotype of male-factor infertility and reflects impaired spermatogenesis rather than ductal obstruction. Microdissection testicular sperm extraction (micro-TESE) is the primary sperm retrieval method, but sperm retrieval rates remain at approximately 40-60% and vary with etiology, genetics, histopathology, surgical expertise, and endocrine ph…

- Developing Potential Endocrine Therapy Controlling Estradiol and DHT by Targeting 17β-HSD7 Against ER+ Breast Cancer. [Journal Article]
- Breast cancer (BC) is the most incidental cancer in women. Patients' living conditions and survival rates have significantly improved with the success of two first-line endocrine therapies of selective estrogen-receptor modulators (SERMs from 1977) and aromatase inhibitors (AIs from 1995). Unfortunately, both therapies have produced significant resistance. AI resistance reaches 50% in metastatic …

- Reassessing Tcf21-iCre for Adult Cardiac Fibroblast Gene Deletion: Limited Recombination Efficiency Following Tamoxifen Injections. [Journal Article]
- Cardiac fibrosis plays a crucial role in heart failure, but research in this field remains challenging due to heterogeneity of cardiac fibroblast populations. In the past years, several Cre-drivers have been proposed for cardiac fibroblast-specific deletion of floxed genes in adult mouse research models. Here, we evaluated whether Tcf21-iCre, a tamoxifen-inducible Cre recombinase (iCre) under the…

- The Role of Sex Hormone Receptors in the Squamous Cell Carcinoma of the Uterine Ectocervix: A Review and Future Directions. [Review]
- Cervical cancer (CxCa) remains a major global gynecological malignancy causally linked to high-risk human papillomavirus (HPV) infection. However, HPV alone is insufficient for carcinogenesis; sex hormones and their receptors serve as essential cofactors. This review aims to synthesize current knowledge on the role of cervical histoarchitecture and hormonal microenvironments in ectocervical squam…

- Design, synthesis, and biological evaluation of selective estrogen receptor modulators (SERMs) as anticancer agents for ER-positive breast cancer. [Journal Article]Bioorg Med Chem. 2026 Aug 11; 142:118763. [Online ahead of print]BM
- Globally, breast cancer stands as one of the primary causes of cancer related deaths. Among the types of breast cancer, estrogen receptor- positive (ER+) breast cancer accounts for the highest incidence and mortality worldwide. The present study emphasizes the design, synthesis, and biological evaluation of a series of novel triphenylethylene (TPE) based selective estrogen receptor modulators (SE…

- Tamoxifen Therapy Is Associated With Altered Intestinal P-Glycoprotein Activity In Vivo. [Journal Article]J Clin Pharmacol. 2026 Aug; 66(8):e70256.JC
- Preclinical studies suggest that tamoxifen (TAM) may interact with the efflux transporter P-glycoprotein (P-gp); however, its effect on intestinal P-gp activity in vivo remains poorly characterized. This study evaluated intestinal P-gp activity in women receiving tamoxifen therapy using fexofenadine (FEXO) as a probe substrate. Sixteen women receiving tamoxifen (20 mg/day for ≥80 days) and 12 hea…

- Hormonal therapies for endometriosis-associated pain: inflammatory limitations and pharmacological challenges. [Review]
- Endometriosis is a chronic inflammatory and estrogen-dependent disease in which pain remains the leading cause of impaired quality of life. Hormonal therapies constitute the cornerstone of medical management and include progestins, combined estrogen-progestin contraceptives, gonadotropin-releasing hormone (GnRH) agonists and antagonists, selective estrogen and progesterone receptor modulators, an…

- Dose-dependent perinatal risks of clomiphene citrate in US IVF cycles: a national cohort study, 2004-2021. [Journal Article]
- CONCLUSIONS: Our findings extend previous findings on the association between CC exposure and adverse perinatal outcomes by demonstrating a dose-dependent relationship.

- Active Peyronie's disease as an early antifibrotic treatment window: a translational review. [Review]Sex Med. 2026 Oct; 14(5):qfag068.SM
- CONCLUSIONS: Active/progressive PD is a defensible setting for early antifibrotic trials. PDE5 inhibitor/SERM therapy is among the best-developed mechanistic candidates, but remains investigational. Clinical adoption requires prospective, randomized, stage-defined studies with standardized curvature assessment, plaque imaging, erectile-function measurement, pain assessment, co-intervention documentation, adverse-event capture, and PD-specific patient-reported outcomes.

- Toremifene-induced bilateral macular crystalline deposits with cystic cavitation: a case report. [Case Reports]Front Med (Lausanne). 2026; 13:1840738.FM
- Tamoxifen-induced retinopathy is well recognized and frequently reported in clinical practice. By contrast, ocular adverse events associated with the structurally similar agent toremifene remain rarely described. We present a case of bilateral visual impairment in a patient receiving long-term oral toremifene citrate for secondary prevention after breast cancer surgery. Ophthalmic evaluation show…

- The tumour microenvironment influences long-term tamoxifen benefit in postmenopausal ER+/HER2- breast cancer patients: a secondary analysis of the randomised Stockholm Tamoxifen (STO-3) trial. [Randomized Controlled Trial]
- CONCLUSIONS: TME composition modulates tamoxifen benefit in postmenopausal ER+HER2- breast cancer. Low immune, intermediate endothelial, and low/intermediate fibroblast abundances are associated with improved benefit from tamoxifen, suggesting that both immune and stromal compartments influence endocrine treatment efficacy.

- [Retinal intoxication with tamoxifen treatment for breast cancer]. [Case Reports]Rev Med Liege. 2026 Jul; 81(7-8):475-481.RM
- Breast cancer is often treated with tamoxifen, a drug that can cause ocular side effects, including toxic retinopathy. Multimodal imaging, particularly optical coherence tomography (OCT), has improved the detection of early lesions such as foveal pseudocysts, thus increasing the prevalence of this complication. We present a case of tamoxifen-induced retinopathy in a patient treated for four years…

- CONCLUSIONS: Raloxifene shows potential as an adjunctive treatment in post- and perimenopausal psychosis, but further high-quality clinical trials are needed to inform practice and develop hormone-informed treatment protocols.
