- A digitally immune-optimized influenza vaccine broadly neutralizes swine and human H1N1 influenza viruses and protects from heterologous challenge. [Journal Article]J Virol. 2026 Oct 06; :e0107326. [Online ahead of print]JV
- Influenza A virus (IAV) zoonotic transmission and continuous evolution in multiple host species heighten the risk of emerging novel strains at the human-animal interface. Digitally immune-optimized hemagglutinin (HA), neuraminidase (NA), and matrix-2 (M2) vaccine antigens were engineered and combined as a single DNA expression construct (DVX-H1N1) to maximize immune responses to human seasonal, p…

- Detection of Influenza D Virus using Reverse Transcription Loop-Mediated Isothermal Amplification. [Journal Article]
- The Orthomyxoviridae family includes influenza D virus (IDV), an emerging pathogen primarily affecting cattle and swine; there is evidence of cross-species transmission and potential zoonotic risk. Although active human infections have yet to be confirmed, high seroprevalence in cattle-exposed populations highlights the need for continued surveillance. We developed and validated a rapid, field-de…

- Molecular Epidemiology and Evolution of Swine Influenza A Viruses, Vietnam, 2020-2024. [Journal Article]
- Swine influenza A viruses (IAV-S) caused the 2009 H1N1 pandemic and pose a future zoonotic and pandemic threat. Vietnam represents a critical hotspot for IAV-S emergence within East and Southeast Asia, with dense swine and human populations and intensive livestock trade. We conducted genomic surveillance of IAV-S in Vietnam during 2020-2024, extending previous surveillance from 2013-2019. We iden…

- PRRSV Fetal Infection in Pigs as a Risk Factor for the Emergence of More Efficient Zoonotic Hosts for Swine-Origin Influenza Virus. [Journal Article]
- Domestic pigs are susceptible to avian, swine, and human influenza A viruses (IAVs). This facilitates co-infections of different IAVs that result in genetic reassortment. In combination with husbandry practices such as high-density farming and frequent human-pig interactions, pigs serve as efficient zoonotic hosts for IAV. Despite significance in zoonotic transmission, factors influencing IAV pat…

- Vaccine-Associated Enhanced Respiratory Disease: From Animal Models to Human Health. [Review]
- Vaccines are a safe and highly effective tool for protecting both humans and animals against infectious diseases. Although vaccines currently in use in humans have robust safety records, historical examples show that some vaccines have resulted in adverse outcomes, including vaccine-associated enhanced respiratory disease (VAERD). VAERD occurs when preexisting vaccine-induced immunity worsens res…

- Substitutions affecting the HA-NA-receptor balance preceded the emergence of the pandemic 2009 H1N1 virus. [Journal Article]
- Swine are a crucial species in influenza A virus (IAV) ecology, capable of supporting replication and consequently evolution of human, avian, and swine strains. They are also the source of the 2009 pandemic H1N1 virus (H1N1pdm09). Understanding how swine IAV breach the species barrier is essential for detection of potentially pandemic viruses, but is often overlooked compared to avian IAV. We hyp…

- Antigenic and structural analysis of the influenza hemagglutinin lateral patch. [Journal Article]
- The lateral patch epitope of the H1 hemagglutinin (HA) was a dominant target of antibodies following exposure to the 2009 pandemic H1N1 virus. However, the conservation and mechanisms of antigenic drift in the lateral patch remain unresolved. Here, we used lateral patch-specific monoclonal antibodies (mAbs) to understand the antigenicity of the lateral patch of human, avian, and swine H1Nx viruse…

- Candidate biomarkers in narcolepsy in H1N1-vaccinated cohorts: A proteomic and transcriptomic study. [Journal Article]Sleep Med. 2026 Dec; 148:109268.SM
- Narcolepsy is a rare neurological disorder caused by the loss of function in neurons producing orexin or hypocretins, disrupting the sleep-wake cycle. In Sweden and Finland approximately 800 patients developed narcolepsy after swine flu (H1N1) vaccination between 2009 and 2010, probably as an autoimmune side effect. The vaccinated cohorts are of great scientific interest owing to their specific e…

- Longitudinal assessment of functional antibodies to a novel influenza virus strain across age groups. [Journal Article]
- Newly emerging influenza viruses pose a constant threat as they encounter a population lacking neutralizing antibodies against the new strain. However, cross-reactive non-neutralizing antibodies (nnABs) may be present and help mitigate disease symptoms through various effector mechanisms, including antibody-dependent cellular cytotoxicity (ADCC). Although nnABs to influenza virus have received mo…

- An enhanced multisegment RT-PCR method for influenza A virus sequencing: Improved performance and reduced preparation time over traditional methods. [Journal Article]J Virol Methods. 2026 Sep 08; 347:115453. [Online ahead of print]JV
- Influenza A viruses (IAVs) remain a major global health threat, affecting both human and animal populations. Whole-genome sequencing is essential for monitoring viral evolution, zoonotic transmission, and emerging variants. However, conventional RT-PCR methods often result in incomplete gene coverage, amplification biases, and reduced sequencing accuracy, particularly in clinical samples. We deve…

- Influenza A virus in Swiss pig herds with respiratory disease: Seasonality and age dependence. [Journal Article]Schweiz Arch Tierheilkd. 2026 Sep; 168(9):143-156.SA
- Influenza A virus (IAV) is an important respiratory pathogen in pigs and poses a zoonotic risk to humans in close contact. While IAV epidemiology has been extensively studied in large-scale production systems, data from Switzerland - characterized by small herds and limited live pig imports - remain scarce. This exploratory nationwide cross-sectional study aimed to assess the association between …

- Cell culture-based manufacturing platforms, already well-established for human influenza vaccines, are increasingly being adopted in the veterinary field as a powerful alternative to traditional embryonated hen's eggs, providing improved scalability and safety. In this study, we established a serum-free suspension culture process using Madin-Darby canine kidney (MDCK) cells for the efficient prod…

- Site-specific microRNA responses in lungs of pigs depend on the host-adaptation of H1N1 influenza A virus. [Journal Article]Virology. 2026 Dec; 624:111053.V
- Cross-species transmission of influenza A virus (IAV) represents a major risk for emergence of new pandemics, however, the host molecular responses invoked by cross-species IAV infection remain incompletely defined. MicroRNAs (miRNAs) are important post-transcriptional regulators of innate immunity, but their role during infection with IAV strains of the same subtype but of different host origin …

- Identification and validation of key host genes associated with porcine H1N1 infection based on integrated machine learning algorithms. [Journal Article]
- CONCLUSIONS: This study combined transcriptomics and multi-machine learning to identify and verify host genes for swine H1N1 infection. SPP1 acts as a stable diagnostic biomarker whose reduced expression correlates with disease progression. Our results reveal new molecular mechanisms of H1N1 pathogenesis and offer a candidate target for swine flu control and zoonotic risk intervention.

- The Important Role of Superoxide Dismutase 2 in Controlling Poxvirus Proliferation and Pathogenicity. [Journal Article]
- Superoxide dismutase 2 (SOD2), a key mitochondrial antioxidant enzyme, is essential for maintaining cellular redox homeostasis by scavenging superoxide radicals. While viruses often induce oxidative stress, the specific role of SOD2 in antiviral defense remains unclear. Here, we report that vaccinia virus (VACV) infection triggers mitochondrial and cellular reactive oxygen species (ROS) and selec…
