- The Organization of the Golgi Structures during Drosophila Male Meiosis Requires the Citrate Lyase ATPCL. [Journal Article]
- During spermatogenesis, the Golgi apparatus serves important roles including the formation of the acrosome, which is a sperm-specific organelle essential for fertilization. We have previously demonstrated that D. melanogaster ATP-dependent Citrate Lyase (ATPCL) is required for spindle organization, cytokinesis, and fusome assembly during male meiosis, mainly due to is activity on fatty acid biosy…

- The role of acroblast formation during Drosophila spermatogenesis. [Journal Article]
- Protein recycling is important for maintaining homeostasis of the Golgi and its cisternae. The Vps54 (Scat) protein, a subunit of the GARP tethering complex, is a central factor in retrograde transport to the trans-Golgi. We found the scat(1) mutant to be male sterile in Drosophila with individualization problems occurring during spermatogenesis. Another typically observed phenotype was the abnor…

- Mutations in Cog7 affect Golgi structure, meiotic cytokinesis and sperm development during Drosophila spermatogenesis. [Journal Article]
- The conserved oligomeric Golgi (COG) complex plays essential roles in Golgi function, vesicle trafficking and glycosylation. Deletions in the human COG7 gene are associated with a rare multisystemic congenital disorder of glycosylation that causes mortality within the first year of life. In this paper, we characterise the Drosophila orthologue of COG7 (Cog7). Loss-of-function Cog7 mutants are via…

- A potential role for the clathrin adaptor GGA in Drosophila spermatogenesis. [Journal Article]
- CONCLUSIONS: In Drosophila we have uncovered a potential role for GGA in the testes of male flies. The gender specific higher expression of GGA, its specific enrichment in testes and its localisation to developing spermatocytes and at the acroblast of spermatids supports a role for GGA function in Drosophila spermatogenesis, even though spermatogenesis still occurs when GGA expression is depleted to <5% of control.

- The Drosophila Cog5 homologue is required for cytokinesis, cell elongation, and assembly of specialized Golgi architecture during spermatogenesis. [Journal Article]
- The multisubunit conserved oligomeric Golgi (COG) complex has been shown previously to be involved in Golgi function in yeast and mammalian tissue culture cells. Despite this broad conservation, several subunits, including Cog5, were not essential for growth and showed only mild effects on secretion when mutated in yeast, raising questions about what functions these COG complex subunits play in t…

- Localization of thiamine pyrophosphatase activity in the golgi apparatus of a mollusc, Helix aspersa. [Journal Article]
- Spermatids of the snail Helix aspersa were studied after fixation in buffered osmium tetroxide and after applying Novikoff and Goldfischer's method (15) for demonstrating thiamine pyrophosphatase (TPPase) activity both with the light and the electron microscope. The appearance of cells in the light microscope after localizing the enzyme is very similar to the appearance after the application of c…

- Acrosome formation in the house cricket. [Journal Article]
- Acrosome formation during spermatogenesis in the house cricket was studied with the electron microscope. In the early spermatid there is a single Golgi body, called the acroblast, which is cup-shaped, the walls being composed of a number of parallel membranes. A pro-acrosomal granule then appears within the acroblast. Next, the granule migrates to the nucleus, where it becomes attached. The acrob…

- Electron microscope studies on the dictyosomes and acroblasts in the male germ cells of the cricket. [Journal Article]
- The dictyosome (Golgi body) in the secondary spermatocyte of the cricket appears in electron micrographs as a duplex structure composed of (a) a group of parallel double-membraned lamellae and (b) a group of associated vacuoles arranged along the compact lamellae in a chain-like fashion. This arrangement of ultramicroscopic structure for the dictyosomes is strikingly comparable to that described …
