- Deficiency of the Lipoprotein Lipase Pathway Modifies the Phenotypic Expression of Genetically Determined Lipoprotein(a). [Journal Article]J Lipid Res. 2026 Oct 05; :101164. [Online ahead of print]JL
- Plasma lipoprotein(a) [Lp(a)] concentrations are primarily genetically determined, but whether disruption of triglyceride-rich lipoprotein metabolism modifies Lp(a) expression is unknown. Familial chylomicronemia syndrome (FCS) provides a human model to investigate this relationship. In the phase 3 Balance trial, 66 participants with genetically confirmed FCS (triglycerides ≥880 mg/dL) underwent …

- Proteomic and Kinetic Analyses Reveal Discordant Apolipoprotein Turnover and Support a Revised Model of Human Lipoprotein(a) Metabolism. [Journal Article]bioRxiv. 2026 Sep 08.B
- Lipoprotein(a) [Lp(a)] is a causal risk factor for atherosclerotic cardiovascular disease composed of apolipoprotein(a) [APO(a)] covalently linked to apolipoprotein B100 (APOB). Although plasma Lp(a) concentrations are largely genetically determined, the mechanisms governing Lp(a) metabolism after particle assembly remain poorly understood. We performed a cross sectional, observational study to d…

- The Roles of Lipoprotein(a) in Atherosclerosis - Minding the Knowledge Gaps. [Review]Curr Atheroscler Rep. 2026 Sep 30; 28(1).CA
- Although the association of lipoprotein(a) (Lp(a)) with coronary heart disease was reported over 60 years ago, and subsequent studies have identified a causal and independent role for Lp(a) in disease, many fundamental unanswered questions persist surrounding the biology of this enigmatic lipoprotein. There remain critical questions surrounding the structure and metabolism of Lp(a) and the unique…

- Multivariate Genome-Wide Association Analysis Identifies Genetic Susceptibility Loci for Metabolic Syndrome in the Korean Genome and Epidemiology Study. [Journal Article]
- Metabolic syndrome (MetS), characterized by a cluster of interrelated metabolic abnormalities including central obesity, elevated blood pressure, dysglycemia, hypertriglyceridemia, and reduced high-density lipoprotein cholesterol (HDL-C), substantially increases type 2 diabetes and cardiovascular disease risk. Conventional genome-wide association studies (GWASs) analyze individual traits or a dic…

- CRISPR/dCas9-induced upregulation of endogenous apolipoprotein A1 and paraoxonase 1 genes reduces the aortic lipid deposits in apoE-/- mice. [Journal Article]
- High-density lipoproteins (HDL) are essential to alleviate the progression of atherosclerosis by mediating reverse-cholesterol transport, antioxidant and anti-inflammatory effects. We aimed to enhance the expression of endogenous HDL components, apolipoprotein A1 (APOA1) and antioxidant enzyme paraoxonase 1 (PON1), and to investigate their athero-protective effects. The CRISPR/dCas9 technology wa…

- Context-Dependent Associations of the APOA1 rs670 Polymorphism with Apolipoprotein A1 and HDL Cholesterol: A Systematic Review and Meta-Analysis. [Systematic Review]
- The APOA1 rs670 (-75 G>A) polymorphism has been implicated in lipid metabolism and susceptibility to metabolic syndrome (MetS). However, individual studies have reported inconsistent associations with circulating apolipoprotein A1 (Apo A1) and lipid parameters, and no meta-analysis has comprehensively evaluated these associations stratified by metabolic disorder status. We therefore quantified th…

- Clinical and functional evidence supporting pathogenicity of a novel APOA5 variant in familial chylomicronemia syndrome. [Journal Article]
- CONCLUSIONS: Measurement of LPL activity demonstrated the functional impact of the APOA5 c.694T>C; p.(Ser232Pro) variant. These findings support reclassification of the variant as likely pathogenic and enable differentiation from multifactorial chylomicronemia syndrome.

- Apolipoprotein(a)-rich electronegative HDL accelerates vascular aging and atherosclerosis by inducing endothelial senescence. [Journal Article]
- The limited benefits of high-density lipoprotein (HDL)-targeted therapies have shifted the research focus to HDL quality; some HDL subtypes may be pro-atherogenic and may counteract the potential benefits of HDL therapies. This study investigated the pathological mechanism of H5, a highly electronegative and compositionally abnormal HDL subfraction characterized by impaired cholesterol efflux. We…

- Antitumor activity of high-density lipoproteins and their use for targeted drug delivery. [Review]Biomed Khim. 2026 Sep; 72(4):255-266.BK
- Blood levels of high-density lipoprotein (HDL) are significantly inversely correlated with the risk of numerous malignancies and overall mortality in most tumor types. This review focuses on the antitumor role of HDL and its main protein component, apolipoprotein A-I (apoA-I). The association of HDL/apoA-I with various cancer types and the probable mechanisms of their antitumor action, including …

- Effect on Lipoprotein(a) Oxidized Phospholipids of Lepodisiran, an Extended-Duration siRNA Targeting Lipoprotein(a). [Journal Article]J Am Coll Cardiol. 2026 Aug 31. [Online ahead of print]JACC
- CONCLUSIONS: Lepodisiran produced sustained reductions in OxPL-apoB and OxPL-apo(a) levels that correlated with Lp(a) lowering. These findings provide additional biological rationale for the ongoing lepodisiran phase 3 cardiovascular outcomes trial, but do not establish clinical benefit. - Site-Specific Apo AI Glycation Impairs HDL Function and Promotes Atherosclerosis in Diabetes. [Journal Article]Circulation. 2026 Sep 22; 154(12):1088-1108.Circ
- CONCLUSIONS: This study provides a comprehensive site-resolved map of apo AI glycation in patients with diabetes and identifies the Apo AI Glycation Index as a glycation signature associated with diabetic CAS and impaired HDL function. Experimental data support a mechanism in which site-specific apo AI glycation contributes to HDL dysfunction and macrophage cholesterol efflux impairment through RAGE-associated NR2C2-LXRα signaling. Glycation-resistant apo AICL preserves HDL function and attenuates atherosclerosis in preclinical models, supporting site-specific apo AI glycation as a mechanistically relevant feature of diabetic atherosclerosis.

- CONCLUSIONS: In patients with high-risk cardiovascular disease undergoing percutaneous intervention, Lp(a), OxPL-related biomarkers, and TCS are influenced by race and gender and correlated with hypercoagulability. These findings suggest novel mechanistic insights into the relationship between Lp(a), OxPL biomarkers, and the propensity for whole-blood clotting and may be relevant in explaining the outcomes of patients in ongoing trials targeting the reduction of Lp(a).
