  • CONCLUSIONS: Repeat-length expansion generates stable, predominantly parallel G4 structures from TTGGG repeats in vitro, identifying expanded plant mitochondrial loci as candidate non-B-DNA-prone regions whose in vivo function remains unresolved. G-quadruplexes (G4s) are non-canonical DNA structures formed by stacked guanine tetrads stabilized by Hoogsteen base pairing and monovalent cations. Although G4s have been extensively studied in nuclear genomes and telomeric DNA, much less is known about G-rich tandem repeats with G4-forming potential in organellar genomes. Here, we investigated exact TTGGG tandem repeats in plant mitochondrial genomes and asked whether repeat expansion can generate local G4-compatible sequence architectures. We first used the exact (TTGGG)4 motif as the shortest TTGGG-derived repeat module compatible with one intramolecular G4-like fold and re-annotated BLAST-positive Viridiplantae records to identify mitochondrion-annotated candidates. Across the curated mitochondrial subset, exact TTGGG tracts formed a repeat-length continuum. The strongest genome-level example was detected in the Zea perennis mitochondrial genome, where a continuous reverse-strand (TTGGG)16 tract was recovered. A complementary G4Hunter analysis of four maize reference mitogenomes recovered the Z. perennis locus as an extended G4-prone region overlapping the (TTGGG)16 tract, whereas no comparable TTGGG-overlapping extended G4Hunter-supported region was detected in the other Zea references examined. Biophysical characterization of synthetic TTGGG-derived oligonucleotides, (TTGGG)4, (TTGGG)8, and (TTGGG)12, revealed robust folding into predominantly parallel G-quadruplex structures, supported by circular dichroism (CD) spectroscopy and native gel electrophoresis. Thermal denaturation experiments further showed high stability under potassium-rich conditions. Together, these results identify expanded TTGGG repeats as an empirical example of G-rich mitochondrial tandem repeats in which repeat expansion increases the number of overlapping minimal G4-compatible modules. More broadly, our data support the view that expanded G-rich repeats can mark local non-B DNA-prone regions in repeat-rich plant mitogenomes, while their actual structural and functional states in vivo remain to be tested experimentally.
  • A modified DNA-barcoding method to identify jumbo squid (Dosidicus gigas) substitution in canned octopus products. [Journal Article]

  • In recent years, the United States Food and Drug Administration (US FDA) has documented several instances where more abundant, and lower priced, jumbo squid (Dosidicus gigas) has been substituted for octopus (family Octopodidae) in a variety of products (e.g. frozen, canned) resulting in the products being misbranded and/or adulterated. FDA typically utilises a short (approx. 655 base pair) fragm…

  • A series of N-heterocyclic carbene (NHC)-stabilized phosphanylgallanes and highly temperature sensitive phosphanylalanes bearing aliphatic and aromatic substituents at the phosphorus atom were synthesized via salt metathesis reactions of IDipp-coordinated (IDipp = 1,3-bis(2,6-diisopropylphenyl)imidazolin-2-ylidene) triel halide precursors with alkali metal phosphanides. The gallium derivatives ID…
  • Infrared spectroscopy of MO(CH4)n (M = Ce, Y; n = 1-3) reveals two distinct adsorption configurations. [Journal Article]

  • Investigation of the interactions of methane with metal oxides is crucial for elucidating absorption configurations at active sites of catalysts, which has proven to be a challenging experimental target due to the difficulty in size selection of neutral systems. In this work, the interactions of methane with rare earth metal oxides are studied by using size-specific infrared-vacuum ultraviolet sp…
  • Structural basis for Mg²⁺-glycine synergy in the tandem glycine riboswitch revealed by cryo-EM. [Journal Article]

  • Riboswitches are non-coding RNA sequences that control cellular processes through ligand binding. Conformational heterogeneity is fundamental to riboswitch functionality, yet this same attribute makes structural characterization of these mRNA elements challenging. Here, we use cryo-electron microscopy to characterize different structural populations of the glycine riboswitch tandem aptamers, and …
  • PNA-ligand conjugates enable site-selective and directional targeting of adjacent G-quadruplexes. [Journal Article]

  • G-quadruplexes (G4s) are non-canonical DNA structures enriched in gene promoters and involved in transcriptional regulation, making them attractive drug targets, albeit difficult to target individually with high specificity due to their structural similarity. In the human c-KIT promoter, three adjacent G4 motifs (c-KIT1, c-KIT*, and c-KIT2) modulate oncogene expression, yet their individual contr…

  • Alpha-thalassemia/mental retardation syndrome X-linked protein (ATRX) is an ATP-dependent chromatin remodeler that performs diverse functions. The mechanism by which ATRX modulates chromatin is unknown. Here, our cryo-electron microscopy (cryo-EM) structure revealed that human ATRX recognizes the nucleosome through its C-terminal regions by embracing and unwrapping ∼18 base pair nucleosomal DNA n…
