Cutaneous siderosis represents a challenging dermatological condition characterized by iron deposition within the skin, resulting in persistent brownish hyperpigmentation that can significantly impact a patient's quality of life. While this condition can arise from various etiologies, intravenous iron infusions remain a common cause due to extravasation and subsequent iron deposition in cutaneous tissues. This case report explores the efficacy of Q-switched 532 nm Nd:YAG laser treatment for cutaneous siderosis, a wavelength selected after demonstrating its superiority over 694 nm ruby and 1064 nm Nd:YAG lasers through direct comparative testing. A 26-year-old woman developed extensive brownish hyperpigmentation following a ferric carboxymaltose infusion. She experienced immediate burning, swelling, and subsequent discoloration extending from her elbow to her wrist. Diagnosis of cutaneous siderosis was confirmed through clinical evaluation. Treatment involved four sessions of Q-switched Nd:YAG laser therapy at a 532 nm wavelength, based on the previous comparative findings. Treatment parameters ranged from 0.6 J/cm[2] to 1.0 J/cm[2] fluence with a 4 mm spot size. Clinical outcomes were assessed using a systematic approach with digital photographs and the Global Aesthetic Improvement Scale (GAIS). Gradual pigment reduction was observed throughout the treatment course, with complete resolution of hyperpigmentation achieved two years after the first treatment session. Q-switched Nd:YAG laser therapy at 532 nm is an effective treatment for cutaneous siderosis resulting from iron infusion extravasation. This case demonstrates the potential for successful removal of iron-induced skin pigmentation with minimal adverse effects, offering a viable solution for patients experiencing this complication.

We aim to discuss the use of laser for the treatment of eyebrow microblading and cosmetic tattoo complications through a review of the literature. Our research question is whether quality-switched or picosecond laser is superior for the removal of eyebrow tattoos. This structured review was conducted using a PubMed search using the search terms "laser tattoo removal" AND "cosmetic tattoo" AND "eyebrow" with the article type filtered to "case reports," "clinical trial," and "randomized controlled trial" ranging from dates 1994-2023. All case reports or series evaluating the effect of laser on eyebrow cosmetic tattooing pigment were included. We summarize the results of 11 studies evaluating the use of laser for cosmetic tattoo removal, with 129 patients treated specifically for eyebrow pigment. Most studies (8/11) report Fitzpatrick skin type or race. Seven studies utilize quality-switched (QS) neodymium-doped yttrium aluminum garnet (Nd:YAG), alexandrite or ruby, three used picosecond (PS) Nd:YAG or alexandrite, and three used carbon dioxide (CO2) laser. We report laser energy, spot size, and pulse duration, as well as treatment outcomes and adverse events. Historically, methods of pigment removal included dermabrasion, cryosurgery, electrosurgery, radiofrequency, infrared light, intense pulsed light, and surgical excision; however, these methods often led to poor cosmetic outcomes including scarring and further dyspigmentation. QS laser treatments provided superior cosmetic outcomes and thus were considered the gold standard treatment option for pigment removal. However, the advent of PS lasers has challenged this given their increased selectivity, lower fluence requirements, and reduction in surrounding thermal damage. Our review demonstrates that PS Nd:YAG is quicker and more effective that QS Nd:YAG in the treatment of eyebrow tattoos. Additionally, the paradoxical darkening seen with QS lasers is less common with PS lasers. We also demonstrate that CO2 laser may be a helpful adjunct to QS or PS laser. This review focuses on Fitzpatrick skin type and race, providing a unique perspective on the use of laser treatment in skin of color, which often poses an additional treatment challenge.

Identification and segregation of citrus fruit with diseases and peel blemishes are required to preserve market value. Previously developed machine vision approaches could only distinguish cankerous from non-cankerous citrus, while this research focused on detecting eight different peel conditions on citrus fruit using hyperspectral (HSI) imagery and an AI-based classification algorithm. The objectives of this paper were: (i) selecting the five most discriminating bands among 92 using PCA, (ii) training and testing a custom convolution neural network (CNN) model for classification with the selected bands, and (iii) comparing the CNN's performance using 5 PCA bands compared to five randomly selected bands. A hyperspectral imaging system from earlier work was used to acquire reflectance images in the spectral region from 450 to 930 nm (92 spectral bands). Ruby Red grapefruits with normal, cankerous, and 5 other common peel diseases including greasy spot, insect damage, melanose, scab, and wind scar were tested. A novel CNN based on the VGG-16 architecture was developed for feature extraction, and SoftMax for classification. The PCA-based bands were found to be 666.15, 697.54, 702.77, 849.24 and 917.25 nm, which resulted in an average accuracy, sensitivity, and specificity of 99.84%, 99.84% and 99.98% respectively. However, 10 trials of five randomly selected bands resulted in only a slightly lower performance, with accuracy, sensitivity, and specificity of 98.87%, 98.43% and 99.88%, respectively. These results demonstrate that an AI-based algorithm can successfully classify eight different peel conditions. The findings reported herein can be used as a precursor to develop a machine vision-based, real-time peel condition classification system for citrus processing.

Melanoma is a metastatic cancer resistant to a wide range of therapies, including standard chemotherapy and radiation therapy, and cannot be treated with existing treatments owing to its intrinsic drug resistance. In terms of convenience and cheap cost of fabrication, one of the novel treatments is using polydopamine-coated iron oxide nanoparticles (IONs@PDA). Iron oxide nanoparticles (IONs) were synthesized (7.36 nm) and coated with polydopamine (15-20 nm). To examine the effect of photothermal ablation in melanoma cells (B16-F10), a Q-switched ruby laser (λ = 694 nm, spot size = 4 mm, output power = 5 J/s) was used. The prepared nanoprobe was applied to mice, and their survival after treatment was evaluated. Then histopathological studies were done on the livers and skins of the treated mice. The nanoparticles absorb the laser, raising the temperature and initiating photothermal treatment, with significant apoptosis (74%) after the 4th time of treatment. Photothermal therapy (PTT) by using IONs@PDA proved to be effective in the treatment of melanoma cells (tumor size of < 2 mm) without side effects. The lifespan of mice was significantly increased in a group of mice post-administered IONs@PDA and laser ablation. The fabricated nanoprobe (IONs@PDA) enhanced the melanoma cell apoptosis in the mice model, and it has promise for the treatment of melanoma (B16-F10) cells using photothermal therapy.

Marine resources are often shared among countries, with some fish stocks straddling multiple Exclusive Economic Zones, therefore understanding the structure of populations is important for the effective management of fish stocks. Otolith chemical analyses could discriminate among populations based on differences in the chemical composition of otoliths. We used otoliths from two deepwater snappers (flame snapper Etelis coruscans and ruby snapper Etelis boweni) to examine the evidence for population structure across six Pacific Island countries using solution-based inductively coupled plasma mass spectrometry (ICP-MS) for otolith core and whole otolith samples and laser ablation ICP-MS (LA-ICP-MS) for core and edge areas of a cross-sectioned otolith. The inter-species comparison of these methods is important as the two species are often managed under the same regulations. For both species, the two methods demonstrated separation among the locations sampled with high classification accuracy. Smaller laser ablation spot size gave greater temporal resolution over the life-history transect. Comparing the early life-history section of the otoliths (i.e., the core), one interpretation is that young fish experienced more uniform environments in the open ocean as larvae than adults, as the elemental fingerprints had greater overlap among multiple locations. LA-ICP-MS methods had some advantages over solution-based ICP-MS and generally better discrimination for the trace elements investigated. There were substantial differences between species, but both methods suggested nonmixing populations at the regional scale. Otolith chemistry can be an effective tool in discriminating variation for deepwater marine species in multispecies fisheries, and edge measurements from LA-ICP-MS provided the greatest resolution. Although caution should be taken in interpreting the results from relatively small samples sizes, otolith chemical analyses could be useful at these spatial scales to investigate population structure. This information on separate or overlapping populations could be used in future regional fishery management plans.

Theory predicts that less fit individuals would disperse more often than fitter ones (Fitness Associated Dispersal, FAD hypothesis). To test this prediction under laboratory conditions, an entire life cycle of Lamium amplexicaule plants and the preferences of its dispersal agent, Messor ebeninus ants, were tracked. Characterization of individual L. amplexicaule plant revealed high variability in spot cover on the surface of the seeds, where less fit plants produce "unspotted seeds" (see Fig. 1 in Introduction). Unspotted L. amplexicaule seeds showed higher variation in germination time and lower germination rate. Moreover, M. ebeninus ants preferably collected these unspotted seeds. Our results show that low fitness L. amplexicaule plants produce seeds with higher potential for dispersal, supporting the FAD hypothesis in a plant-animal system.

Dried blood spots have been used as alternatives to traditional plasma and serum samples. We have now developed new devices, named volumetric absorptive paper disc (VAPD) and mini-disc (VAPDmini), to collect accurate volumes of dried blood spots in a simple manner and without the need for additional instruments. VAPD consists of a filter paper disc and a filter paper sheet with holes slightly larger than the disc. The disc is fixed in one such hole without direct contact with the filter sheet. VAPDmini is a scaled-down version of the same device. When several drops of whole blood are applied, the disc becomes saturated and any excess sample is absorbed by the surrounding filter sheet. Accuracy and precision of sampling were assessed by determining the levels of clozapine and its metabolites as target analytes by liquid-liquid extraction and high-performance liquid chromatography with coulometric detection. In addition, differences in analyte recovery were within ±15% for all analytes in samples with 30-60% hematocrit, suggesting that VAPD and VAPDmini are insensitive to hematocrit for the analytes tested. The devices were also validated for analyte concentrations in the range 50-1000 ng/mL, and the limit of detection and lower limit of quantification were 5-17 ng/mL and 15-51 ng/mL, respectively. Intra- and inter-day precision ranged from 3% to 13%, whereas accuracy ranged from a -14% to 12% bias. Analytes were stable in the devices for at least 2 weeks at room temperature. Collectively, these results indicate that sampling using VAPD and VAPDmini is comparable to conventional hole punch sampling of entire dried blood spots, even for samples obtained from patients treated with clozapine. Importantly, the devices were also found to be suitable for sample self-collection.

Cercospora leaf spot (CLS; Cercospora beticola) is the most important foliar disease affecting table beet. Epidemics occur annually and fungicides extend the survival of foliage to enable mechanized harvest. However, a high frequency of strobilurin-resistant C. beticola isolates necessitates the identification of fungicides with different modes of action for tactical rotation. There is also substantial demand for organically produced table beet, for which synthetic fungicides are prohibited. Five small-plot, replicated field trials were conducted over two years to evaluate conventional and Organic Materials Review Institute (OMRI)-listed products for CLS control in table beet cv. Ruby Queen at Geneva and Ithaca, New York. Benzovindiflupyr + difenoconazole significantly reduced temporal disease progress (measured by the area under the disease progress stairs; AUDPS) by 86.7 to 97.3% compared with nontreated plots, and mean survival time of leaves was significantly extended. The demethylation inhibitor, propiconazole, also provided significant disease control in two trials in 2016. Disease severity in plots treated with succinate dehydrogenase inhibitors (boscalid, fluxapyroxad + pyraclostrobin, and penthiopyrad) was significantly decreased compared with nontreated plots but less than other fungicides. Efficacious fungicides significantly increased the dry weight of foliage but did not significantly affect the dry weight of roots, and root shoulder diameter. The enhanced longevity of leaves and increased dry weight of foliage may extend opportunities for mechanized harvesting without deleteriously affecting root yield parameters which are strictly regulated for the processing markets. In two trials, copper octanoate + Bacillus amyloliquefaciens strain D747 (as Cueva + Double Nickel LC) resulted in significantly improved disease control in comparison with application of either product alone and provided comparable and reproducible disease control equivalent to conventional fungicides at both locations. The implications of these findings for CLS control in conventional and organic table beet production systems are discussed.

Chronic K(+) deficiency can cause hypokalemic nephropathy associated with metabolic alkalosis, polyuria, tubular dilatation, and tubulointerstitial injury. However, effects of acute K(+) deficiency on the kidney remained unclear. This study aimed to explore such effects by evaluating changes in levels of proteins in renal tubular cells during acute K(+) deficiency. MDCK cells were cultivated in normal K(+) (NK) (K(+)=5.3 mM), low K(+) (LK) (K(+)=2.5 mM), or K(+) depleted (KD) (K(+)=0 mM) medium for 24 h and then harvested. Cellular proteins were resolved by two-dimensional gel electrophoresis (2-DE) and visualized by SYPRO Ruby staining (5 gels per group). Spot matching and quantitative intensity analysis revealed a total 48 protein spots that had significantly differential levels among the three groups. Among these, 46 and 30 protein spots had differential levels in KD group compared to NK and LK groups, respectively. Comparison between LK and NK groups revealed only 10 protein spots that were differentially expressed. All of these differentially expressed proteins were successfully identified by Q-TOF MS and/or MS/MS analyses. The altered levels of heat shock protein 90 (HSP90), ezrin, lamin A/C, tubulin, chaperonin-containing TCP1 (CCT1), and calpain 1 were confirmed by Western blot analysis. Global protein network analysis showed three main functional networks, including 1) cell growth and proliferation, 2) cell morphology, cellular assembly and organization, and 3) protein folding in which the altered proteins were involved. Further investigations on these networks may lead to better understanding of pathogenic mechanisms of low K(+)-induced renal injury.
