- Machine-learning-guided library screening and drug resistance profiling of HIV-1 protease inhibitors. [Journal Article]J Mol Graph Model. 2026 Nov; 148:109524.JM
- HIV-1 protease inhibitors are central to antiretroviral therapy, yet their long-term efficacy is undermined by the rapid emergence of drug-resistant variants. Although virtual screening of small-molecule libraries and resistance prediction have each been used to guide inhibitor design, there is still no integrated framework that jointly optimizes intrinsic potency and robustness to protease mutat…

- Effects of New P2X7R Antagonists on Retinal Inflammatory Degenerative Conditions. [Journal Article]
- Purinergic receptor (P2X7R), an adenosine triphosphate (ATP) gated nonselective cation channel, is an emerging target for several neurodegenerative conditions, including retinal degeneration. In particular, P2X7R has an established role in multiple inflammatory and immune responses, being implicated in the formation and activation of NLRP3 inflammasome leading to inflammatory priming and release …

- Deciphering virus host interaction analysis to unravel core virus network signatures and computational drug repurposing for cervical cancer. [Journal Article]Comput Biol Med. 2026 Jan 01; 200:111395.CB
- Cervical cancer (CC) is a major contributor to cancer-related deaths. Human papillomavirus (HPV) genotypes, especially HPV16 and HPV18, are the primary causes of CC-related infections. Although the oncogenic potential of HPV is well established, the molecular mechanisms underlying cancer initiation and persistence remain unclear. In this study, the common proteins between the virus-host interacti…

- Reviewing the Developing Significance of the Serine Protease PRSS23. [Review]
- The serine protease 23 (PRSS23) is a highly conserved member of trypsin-like serine proteases, which are associated with numerous essential processes, including digestion, blood coagulation, fibrinolysis, development, fertilization, apoptosis, and immunity. Original reports on PRSS23 unfolded not earlier than 2006 when a molecular biology study characterized and described PRSS23 as an ovarian pro…

- Synthesis and antiviral activity of new 4-substituted Oxycoumarins against SARS and HSV-2 viruses: Mechanistic and molecular modeling study. [Journal Article]Bioorg Chem. 2025 Sep; 164:108845.BC
- A new series of 4-substituted oxycoumarin scaffolds was designed, synthesized, and evaluated for antiviral activity against both SARS-CoV-2 and HSV-2 viruses. Results indicated that compounds 3a, 4a, 4d, and 6d were found to be the most active members against SARS-CoV-2 with IC50 values of 11.17-16.65 μg/mL, with respect to acyclovir (IC50 15.94 μg/mL). On the other hand, compounds 3a, 4a, 6b, 6c…

- Antiretroviral medications for pediatric use in the context of the Unified Health System: are there therapeutic gaps in Brazil? [Journal Article]
- CONCLUSIONS: The comparison between the national and international essential medicines lists highlights the greater variety of pediatric antiretrovirals in Brazil but underscores the need for more child-friendly formulations. The implementation of protocols for the manipulation and fractionation of solid pharmaceutical forms is urgent in order to improve treatment adherence and therapeutic efficacy.

- Repurposing of approved drugs towards Nipah virus treatment: an in silico docking, molecular dynamics simulation and a MM/GBSA approach. [Journal Article]
- The Nipah virus (NiV) is an emerging zoonotic pathogen that causes severe febrile encephalitis, transmitted from infected animals to humans. Since its initial outbreak in Malaysia, subsequent epidemics have occurred across Asia, including India, with mortality rates ranging from 40 to 75%. There are no approved vaccines, or antiviral treatments currently exist for NiV infections. In this study, m…

- Tipranavir analogs as antiviral agents: Design, synthesis, in vitro, and in silico study of new SARS-CoV-2 main protease inhibitors. [Journal Article]Bioorg Chem. 2025 Aug; 163:108624.BC
- The current study aims at developing non-peptidic new antiviral hits against main protease (M[pro]) of SARS-COV2 using the non-peptidic M[pro] inhibitor tipranavir (TPV) as a starting point. In vitro protease inhibition assay of the targeted compounds indicated that compound 6f and its cyclized triazole analogue 7f were the most active in the present study with IC50 values of 6.48 and 7.38 μM, re…

- Total Synthesis of Tipranavir Based on Iridium-Catalyzed Asymmetric Allylic Substitution of Dihydropyranone. [Journal Article]Org Lett. 2024 Nov 29; 26(47):10114-10118.OL
- An efficient and highly enantioselective synthesis of tipranavir is realized based on an iridium-catalyzed asymmetric allylic substitution. High yield and diastereoselectivity (>20:1), as well as excellent enantioselectivity (99% ee), were obtained for the key intermediate through direct asymmetric alkylation reaction of dihydropyranone with allylic tert-butyl carbonate. Anti-AIDS drug of tiprana…
