- Age-stratified safety signal mining of ADHD medications in pediatric populations using the FAERS database (2014-2024): a pharmacovigilance study using multi-method disproportionality analysis. [Journal Article]
- CONCLUSIONS: Distinct age-specific safety signals emerged for ADHD medications, with dextroamphetamine-PRES warranting targeted pharmacovigilance attention.

- Caudal-Threaded Versus Direct Lumbar/Thoracic Epidural Catheters in Small-Weight Infants-A Single-Center Retrospective Observational Study. [Journal Article]
- Background: Continuous epidural analgesia is widely used for postoperative pain management in small infants. Epidural catheters may be inserted via a caudal-threaded approach or by direct lumbar/thoracic placement; however, comparative data on placement success, clinical use, postoperative analgesic outcomes, and complications remain limited. This study compared these two epidural techniques in s…

- Mosaic Trisomy 14 with Severe Short Stature: A Case Report. [Case Reports]
- Mosaic trisomy 14 is a rare chromosomal anomaly with a broad phenotypic spectrum. We report a 10-year-3-month-old girl with severe short stature, developmental delay, and repaired patent ductus arteriosus. An incompletely documented combined insulin-clonidine stimulation test yielded a peak GH concentration of 6.85 ng/mL and was not considered sufficient to establish growth hormone deficiency. Ka…

- A Clonidine-Associated α-Adrenergic Hypothesis in Sleep Bruxism Pharmacotherapy: An Evidence-Anchored Target Analysis. [Journal Article]J Oral Rehabil. 2026 Sep 22. [Online ahead of print]JO
- CONCLUSIONS: This efficacy-anchored analysis generated a clonidine-associated α-adrenergic, arousal-related pharmacological signal within the limited available evidence. The findings are more consistent with a central arousal-related hypothesis for SB pharmacotherapy than with a single motor-target or dopamine-driven treatment model, but require confirmation in contemporary PSG-based clinical studies and real-world patient populations.

- Safety and tolerability study of clonidine for sedation in mechanically ventilated adult patients. [Journal Article]
- CONCLUSIONS: Clonidine was well tolerated within the studied dose range when used as an adjunct to standard sedation in our cohort of non-bradycardic, mechanically ventilated, critically ill adults. No clear dose-dependent increase in predefined adverse events was observed within the studied dose range. These findings provide support for further prospective evaluation of clonidine at doses up to 1800 µg/day in future efficacy studies.

- Diagnostic accuracy of growth hormone stimulation tests for growth hormone deficiency following radiotherapy involving the hypothalamic-pituitary axis: protocol for a systematic review and planned meta-analysis. [Journal Article]
- Radiotherapy is part of the treatment for many intracranial tumours, head and neck cancers and haematological malignancies. In these cases, the hypothalamic-pituitary (HP) axis is often included within the radiation field and can be damaged as an organ at risk. As survival improves, an increasing number of patients are living with late endocrine complications, particularly growth hormone deficien…

- Postoperative Analgesic Efficacy of Extended Opioid-free Anesthesia Regimen with Intravenous Lignocaine versus Morphine Following Major Head and Neck Surgeries. [Journal Article]Ann Afr Med. 2026 Nov 01; 25(6):1711-1715.AA
- CONCLUSIONS: Intravenous lignocaine significantly reduced inadequate analgesia and need for rescue medications with optimal hemodynamics in the immediate postoperative period following major head and neck surgeries. Further large-scale, prospective, randomized controlled trials are warranted.

- StatPearls: Deadly Single Dose Agents [BOOK]StatPearls. StatPearls Publishing: Treasure Island (FL).BOOK
- Several studies have supported Dr Gideon Koren’s 1993 landmark article, “Medications Which Can Kill a Toddler with One Tablet or Teaspoonful.” Although each study has its own merits, most focus solely on pediatric overdoses, and few provide an exhaustive reference list for real-time evaluation of ingestions in children and adults. The following xenobiotics warrant special attention because of the…
- A Novel Multiple Sensory Nerve Block Combination Using Ultrasound Guidance in Knee Arthroplasty: A Randomized Clinical Trial. [Randomized Controlled Trial]A A Pract. 2026 Sep 01; 20(9):e02274.P
- CONCLUSIONS: No differences were observed in pain scores between the eight-nerve block combination and LIA. Secondary outcomes revealed a reduction in 48-hour opioid consumption and a modestly shorter hospital stay with nerve block compared to LIA.

- Practical guide to intrathecal drug delivery: short-term and long-term management and complications. [Review]Reg Anesth Pain Med. 2026 Sep 08; 51(9):1065-1087.RA
- Intrathecal drug delivery (IDD) is an established therapy for patients with refractory pain across cancer and non-cancer populations. By enabling targeted spinal drug delivery, IDD can provide durable symptom control at substantially lower doses than systemic therapy, often reducing systemic medication burden and treatment-limiting adverse effects. Despite decades of clinical experience and suppo…

- Hallucinogen Persisting Perception Disorder in a Young Adult Case Report. [Journal Article]
- Hallucinogen persisting perception disorder (HPPD) is a rare psychiatric diagnosis encompassing a range of perceptual disturbances caused by the lingering effects of hallucinogens. There are two major subtypes of the disorder: HPPD I and II. HPPD II is considered a more severe form that is generally longer-term, distressing, and requiring more intense treatment. In the limited available literatur…

- Prevalence, clinical features and emergency department blood pressure management of hypertensive emergencies and uncontrolled hypertension: a retrospective multi-centre study. [Journal Article]
- Very heterogeneous data have been published concerning prevalence and clinical management of Hypertensive Emergencies (HE) and Uncontrolled Hypertension (UH). The aim of our study was to characterize HE and UH patients evaluating: (i) Blood Pressure (BP) management in Emergency Department (ED), the pharmacological treatment and its adherence to guidelines recommendation; (ii) variables associated…

- Reinstatement of escitalopram and clonidine administration reverses the cessation-induced enhanced activation of rat norepinephrine and serotonin neuronal firing activity. [Journal Article]J Psychopharmacol. 2026 Sep 01; :2698811261478602. [Online ahead of print]JP
- CONCLUSIONS: These results show that abrupt cessation of the SRI escitalopram resulted in robust overactivation of both NE and 5-HT neurons. Normalization of the SRI cessation-induced overactivation of NE and 5-HT neurons by the acute inhibition of the 5-HT reuptake transporter is consistent with the rapid relief of discontinuation symptoms observed in patients resuming their SRI. A similar effect of clonidine on NE and 5-HT neuronal firing would support its use in managing discontinuation symptoms seen after SRI cessation.
- Adjunct Medications for Single-Injection Pediatric Regional Blocks: Implications for Peripheral Nerve Blocks. [Review]
- Single-injection peripheral nerve blocks (PNBs) provide excellent perioperative analgesia in children but are limited by finite duration. Pediatric populations were excluded from the major adult adjunct reviews. This narrative review synthesizes the evidence for adjuncts added to local anesthetics for single-injection PNBs and caudal blocks in patients aged 0 to 18 years, including dexamethasone,…
