- Background and objective Antipsychotic-associated metabolic abnormalities, including weight gain, dyslipidemia, hyperglycemia, obesity, and metabolic syndrome, vary among drugs. Receptor pharmacology offers mechanistic clues, while transcriptome-wide association studies identify genetically regulated expression signals linked to lipid traits. These evidence streams are usually evaluated separatel…

- Impact of nutritional factors on the pharmacokinetics of antipsychotics - a systematic review. [Systematic Review]Psychiatry Res. 2026 Oct; 364:117332.PR
- CONCLUSIONS: Nutritional factors are a significant source of PK variability in antipsychotic treatment. The results underscore the necessity of standardized administration guidelines (e.g., "take with food" for ziprasidone or "fasted" for amisulpride) to ensure predictable plasma levels and optimize therapeutic outcomes in clinical practice.

- CONCLUSIONS: Our findings are consistent with previous NMAs and current guidelines, expanding the current knowledge base while concurrently appraising different drugs, doses, and age groups.

- Heterogeneity and concentration-dependence of ionic channel mechanisms underlying ventricular repolarization prolongation induced by atypical antipsychotics. [Journal Article]Toxicol Appl Pharmacol. 2026 Oct; 515:117973.TA
- Atypical antipsychotics (AAPs)-induced QT interval prolongation represents a major clinical safety challenge. Traditional mechanisms based solely on hERG (IKr) channel blockade cannot fully explain the heterogeneity of clinical phenotypes and severe arrhythmias during overdose. This study systematically evaluated the effects of representative AAPs at therapeutic and toxic concentrations on cardio…

- Cardiac Safety Profile of Ziprasidone: A Real-World Pharmacovigilance Study Using FAERS and Network Toxicology. [Journal Article]Curr Neuropharmacol. 2026 Jul 09. [Online ahead of print]CN
- CONCLUSIONS: The findings from FAERS pharmacovigilance analysis suggest a potential association between ziprasidone exposure and cardiac adverse events. Toxicological network analysis further indicated that the calcium signaling pathway may be involved in the underlying molecular mechanisms.

- Cardiovascular Effects of Droperidol: A Clinically-Focused Review for the Consultation-Liaison and Emergency Psychiatrist. [Review]J Acad Consult Liaison Psychiatry. 2026 Jul 03. [Online ahead of print]JA
- CONCLUSIONS: Despite practical advantages for agitation, the use of droperidol is associated with moderate- to high-risk QTc prolongation, comparable to ziprasidone and greater than intravenous haloperidol. Although droperidol use does not appear to be associated with an increased risk for arrhythmia, alternative agents may be preferred in patients with other risk factors for arrhythmias. Consultation-liaison and emergency psychiatrists should be familiar with these risks.

- Pharmacological and non-pharmacological interventions for altered consciousness and agitation following paediatric acquired brain injury: a scoping literature review. [Review]Disabil Rehabil. 2026 Jun 26; :1-23. [Online ahead of print]DR
- CONCLUSIONS: Overall, the included studies yielded mixed findings. This review highlights the paucity of high-quality experimental studies investigating the use of pharmacological and non-pharmacological approaches to managing altered consciousness and agitation for children following ABI. Further research is needed to determine their efficacy and inform recommendations to guide clinical practice.

- BARD1 phase separation orchestrates a repair hub by enriching XRCC5 to drive chemoresistance in glioma. [Journal Article]Life Sci. 2026 Sep 15; 401:124539.LS
- Temozolomide (TMZ) resistance is a major challenge in glioblastoma (GBM). The role of BRCA1-associated RING domain protein 1 (BARD1), a DNA damage response protein, in GBM and its potential link to liquid-liquid phase separation (LLPS) remain unclear. Bioinformatics analysis of TCGA, CGGA, and GEO datasets revealed that high BARD1 expression correlates with poor prognosis in GBM. Functional studi…

- Risk of extrapyramidal symptoms associated with antipsychotic drugs in children and adolescents: a systematic review and meta-analysis of double-blind randomised placebo-controlled trials. [Journal Article]
- CONCLUSIONS: Antipsychotic drugs are associated with EPS in approximately 1 out of 7 C&A with a more than twofold increased risk compared with placebo. The highest risk estimates were observed for ziprasidone, risperidone, and aripiprazole. Careful risk-benefit assessment, tailored prescribing, and systematic monitoring of EPS in routine paediatric clinical practice are warranted.

- Antipsychotic selection in combination with electroconvulsive therapy for acute schizophrenia: differential efficacy and safety profiles from a real world study. [Journal Article]
- CONCLUSIONS: In this observational study of patients with acute phase schizophrenia, the comparative effectiveness and tolerability of antipsychotic medications combined with ECT appeared to differ depending on the specific agent used. These preliminary observations require confirmation in randomized controlled trials before guiding individualized treatment selection.

- Multi-objective optimization in population pharmacokinetic model selection and optimization: application of NSGA-II in pyDarwin. [Journal Article]
- The selection of a "good" model usually involves a combination of objective and subjective criteria. Although many aspects of model quality can be expressed numerically, certain desirable characteristics remain difficult-or even impossible-to quantify precisely. Multi-objective optimization (MOO) provides a systematic way to handle this challenge by explicitly incorporating and balancing both obj…

- Diagnosis and Medication Treatment of Schizophrenia in Adolescents. [Review]
- Schizophrenia is a chronic mental health condition that most commonly develops in the third decade of life, but may emerge during adolescence, although in rare cases it can also present in childhood. Pre-adult onset is associated with substantial morbidity, functional impairment, and long-term disability. Accurately diagnosing schizophrenia in youth is particularly challenging, as its symptoms fr…

- Seizures Risk Related to Atypical Antipsychotics: A Disproportionality Analysis Based on the FDA Adverse Event Reporting System (FAERS). [Journal Article]Alpha Psychiatry. 2026 Apr; 27(2):44501.AP
- CONCLUSIONS: This large-scale real-world analysis suggests that clozapine, quetiapine, olanzapine, aripiprazole, and particularly ziprasidone are associated with higher reporting signals for seizures compared with risperidone, paliperidone, and lurasidone. The inverse correlation with the 5-HT1A/D2 ratio suggests a potential pharmacodynamic mechanism. Clinicians should consider these differential risks, especially when prescribing for patients with predisposing factors, and maintain vigilance for specific seizures types. These findings warrant further validation through prospective studies and clinical causality assessments.

- Risk factors for drug-related gestational diabetes mellitus: a real-world pharmacovigilance study based on the FAERS database. [Journal Article]
- CONCLUSIONS: This study identified 12 drugs that may be independent risk factors for GDM, including quetiapine, aripiprazole, olanzapine, prednisolone, venlafaxine, risperidone, escitalopram, clozapine, mirtazapine, ziprasidone, rosuvastatin, and trazodone. The established predictive model has certain clinical value, and the potential mechanisms by which antipsychotic drugs may lead to GDM were further analyzed. These results provide potential clues for the early identification of GDM and early warning of suspected drugs, and offer a reference for further exploration of its pathogenesis.
