- Safety and efficacy of guanabenz in early-childhood onset vanishing white matter: primary analysis of a single-arm, phase 1/2 trial. [Clinical Trial, Phase II]Lancet Neurol. 2026 Sep; 25(9):818-829.LN
- CONCLUSIONS: Guanabenz treatment had an acceptable and manageable safety profile and, after the initial phase, was well tolerated in children with vanishing white matter. Treated patients with disease onset at age 6 years or younger, who were still ambulant at baseline, had a significantly lower risk of losing the ability to walk with support than did matched historical controls, although this was a non-randomised comparison. The disease-modifying effect of guanabenz should be confirmed in a long-term extension study.

- Pde4 mediates MHCII expression in oligodendroglia. [Journal Article]
- Chronic psychosocial stress is a major precipitant of Major Depressive Disorder (MDD), yet the glial mechanisms that translate sustained stress into maladaptive myelin and immune changes remain unclear. Using chronic social defeat stress and single-nucleus RNA sequencing of anterior medial PFC (mPFC) oligodendroglia, we identified a mature-oligodendrocyte cluster almost exclusively from stress-su…

- The neuroprotective effect of guanabenz combined with α-lipoic acid in the hSOD1-G93A amyotrophic lateral sclerosis model. [Journal Article]Brain Res Bull. 2026 Jun 01; 239:111890.BR
- Amyotrophic lateral sclerosis (ALS) is a fatal neurodegenerative disease, and although its pathogenesis is not yet clear, the multifactorial mechanisms that affect motor neuron death are intertwined, exacerbating the disease. Here, we explore the effectiveness and mechanism of a combination medication that combines guanabenz with α-lipoic acid in an in vitro as well as in vivo model of ALS. In th…

- The discovery of monoamine oxidase inhibitors: virtual screening and in vitro inhibition potencies. [Journal Article]
- The monoamine oxidase (MAO) enzymes are mitochondrial flavoenzymes that catalyse the oxidative deamination of neurotransmitter amines such as serotonin, norepinephrine and dopamine. Inhibitors of the MAOs are well-known antidepressant and antiparkinsonian agents, and act by reducing MAO-mediated metabolism of neurotransmitters in the brain. The present study attempted to identify compounds that i…

- Pharmacological Enhancement of Integrated Stress Response Confers Protection in Calcific Aortic Valve Disease. [Journal Article]
- Previous studies have shown that Nox4 activates eIF2α/ATF4 signaling during the integrated stress response (ISR) and protects heart injury. However, their roles in calcific aortic valve disease (CAVD) remain unclear. Here, we show that both ATF4 and Nox4 are up-regulated in porcine aortic valve interstitial cells (AVIC) and in human aortic valves with CAVD. Nox4 knockdown promotes while Nox4 over…

- Consensus-Based Expert Recommendations for Diagnosis and Clinical Management of Vanishing White Matter. [Review]
- Vanishing white matter (VWM) is a rare disorder, characterized by degeneration of CNS white matter, clinically often exacerbated by stressors such as fever and minor head trauma. VWM is caused by biallelic pathogenic variants in the EIF2B1-5 genes, causing reduced activity of eukaryotic initiation of translation factor 2B, resulting in dysregulation of the integrated stress response (ISR). New sc…

- Endogenous Repair in Vanishing White Matter. [Journal Article]Ann Neurol. 2026 Mar; 99(3):737-747.AN
- Vanishing white matter is a leukodystrophy with remarkable regional variation in disease severity. The cerebral and cerebellar white matter chronically degenerates, while stress-induced episodes of rapid neurological deterioration coincide with the appearance of acute focal lesions in the deep gray structures and brainstem. With clinical recovery, these acute lesions spontaneously improve or comp…

- Critical Functional Domains in Pediatric Onset TUBB4A-Related Leukodystrophy: A Clinical and Caregiver's Perspective. [Journal Article]
- CONCLUSIONS: Through standardized surveys and outcome assessments, we demonstrate the severe impact of TUBB4A-LD on the quality of life of affected subjects and their caregivers, who experience impairments in motor and expressive language abilities. These results will inform the design of future patient-centric clinical trials.

- Myelin-water imaging and multi-shell diffusion-weighted imaging in adults with adrenoleukodystrophy. [Journal Article]
- The pathophysiology of X-linked adrenoleukodystrophy (ALD) is not well-understood. New quantitative MRI (qMRI) sequences, such as myelin-water imaging (MWI) and multi-shell diffusion-weighted imaging (DWI), are non-invasive techniques that can investigate microstructural abnormalities in the brain. Using these techniques, this study investigated abnormal white matter of cerebral ALD (cALD) lesion…

- The impact of vanishing white matter on unaffected family members. [Journal Article]
- CONCLUSIONS: Mothers and partners indicate a significant and consistent reduction in their quality of life on standardized questionnaires. Qualitative interviews revealed in-depth details of VWM's impact on all family members. Improved healthcare communication, symptom management resources, and support networks are essential for alleviating VWM's impact on families. This study emphasizes the importance of tailored approaches to supporting family members of VWM patients and enhancing their quality of life.

- Literature data-based de novo candidates for drug repurposing. [Journal Article]
- BACKGROUND: Drug repurposing offers a promising strategy for drug discovery. Drug repurposing involves identifying new therapeutic indications for existing, marketed drugs, thereby reducing the risks, costs, and time typically required for drug development. Various methods exist for drug repurposing, including high-throughput screening of drug compound libraries, computation in silico approaches,…
