- Rilzabrutinib, an oral Bruton tyrosine kinase inhibitor, in patients with warm autoimmune haemolytic anaemia: a prospective, single-arm, multicentre, phase 2b safety and clinical activity study (LUMINA 2). [Clinical Trial, Phase II]Lancet Haematol. 2026 Oct; 13(10):e698-e709.LH
- CONCLUSIONS: To our knowledge, this is first clinical study of a Bruton tyrosine kinase inhibitor, rilzabrutinib, in warm autoimmune haemolytic anaemia. Rilzabrutinib showed robust overall and durable haemoglobin response and favourable safety in patients with previously treated warm autoimmune haemolytic anaemia. Rilzabrutinib has the potential to treat the underlying pathophysiology of warm autoimmune haemolytic anaemia through multi-immune modulation, induce a robust and sustained haemoglobin response, and provides an opportunity to decrease or stop corticosteroid use.

- CONCLUSIONS: ARPI doublets demonstrated superior PFS but similar OS compared with docetaxel doublet, with no differences among individual ARPIs. Patients receiving ARPI doublets were less likely to progress to mCRPC or initiate new treatment.

- Longitudinal liquid biopsy and matched radiomic analysis indicated resistance mechanisms of patients with biliary tract cancer undergoing systemic treatment. [Journal Article]Clin Cancer Res. 2026 Oct 01. [Online ahead of print]CC
- CONCLUSIONS: Liquid biopsy identified potential primary (MET SNVs/ERBB2 CNVs) and secondary (EGFR/MET CNVs) resistance mechanisms to platinum-based SACT. Radiomics provided an early correlation with molecular evolution.

- Malignancy-Associated Microangiopathic Hemolytic Anemia Mimicking Thrombotic Thrombocytopenic Purpura in a Patient With Metastatic Breast Cancer: A Case Report. [Case Reports]Cureus. 2026 Aug; 18(8):e115520.C
- Microangiopathic hemolytic anemia (MAHA) is a form of Coombs-negative hemolysis characterized by mechanical fragmentation of erythrocytes, producing schistocytes on the peripheral blood smear. When accompanied by thrombocytopenia and neurological features, the presentation closely resembles thrombotic thrombocytopenic purpura (TTP), a hematological emergency requiring urgent plasma exchange. Howe…

- EZH2-dependent phenotypic plasticity reveals TTP-associated and mTORC1 therapeutic vulnerabilities in lethal prostate cancer. [Journal Article]Cell Rep Med. 2026 Sep 30; :103083. [Online ahead of print]CR
- Phenotypic plasticity is a recognized mechanism of therapeutic resistance in prostate cancer (PCa); however, current knowledge of its underlying drivers and therapeutic interventions remain limited. Using genetically engineered mouse models (GEMMs) devoid of Pten and Rb1, we previously demonstrated the chromatin reprogramming factor enhancer of zeste homolog 2 (EZH2) as a regulator of alternative…

- Current standards and novel concepts in thrombotic thrombocytopenic purpura. [Review]Front Med (Lausanne). 2026; 13:1806030.FM
- Thrombotic thrombocytopenic purpura (TTP) is a rare and rapidly life-threatening thrombotic microangiopathy. Disseminated microvascular thrombosis and resulting ischemia are the critical events determining outcome, if left untreated. The pathogenic hallmark is a severe deficiency of the plasma metalloprotease ADAMTS13, resulting in impaired cleavage of ultra-large von Willebrand factor (vWF) mult…

- Efficacy and safety of T-DXd versus Other HER2-targeted therapies in second-line and later settings for HER2-positive metastatic breast cancer: A Bayesian network meta-analysis. [Journal Article]
- CONCLUSIONS: Within the connected overall networks, T-DXd showed a favorable survival profile and the highest confirmed ORR, although its survival ranking was not estimable in the second-line-only OS or Asia-dominant survival networks. Its increased ILD/pneumonitis risk relative to T-DM1 is clinically important. Indirect survival comparisons and sparse safety networks require cautious interpretation.

- CONCLUSIONS: The purpose of the clinical practice guideline is to improve the acute care and interdisciplinary management of TTP. Further research is needed on the use of recombinant ADAMTS13 and on the definition of uTTP (TTP in the absence of causative antibodies or genetic mutations).

- CONCLUSIONS: The patient-specific TTP with screw fixation improves ankle stability in most loading directions, potentially compensating for ligament insufficiency. However, greater plantarflexion instability necessitates post-operative measures. These findings provide biomechanical evidence that fixation-enhanced TTP designs can improve therapeutic outcomes.

- Intracranial Pressure Estimation Using B4C Device: A Systematic Review on the Validation Studies. [Review]
- Intracranial hypertension (IH) is a frequent and serious complication of acute brain injury, strongly influencing morbidity and mortality. While invasive intracranial pressure (ICP) monitoring is standard, it has limitations. Intracranial compliance (ICC) offers additional insight into the brain's ability to handle changes in volume. Noninvasive ICP waveform analysis, such as the peak 2 (P2)/peak…

- Unveiling the Prognostic Power of Circulating Tumor DNA in Multiple Myeloma: A Systematic Review and Updated Meta-Analysis. [Systematic Review]
- Circulating tumor DNA (ctDNA) serves as a non-invasive liquid biopsy technique, offering potential utility in the prognostic evaluation of multiple myeloma (MM). In this study, we performed a systematic review and updated meta-analysis to analyze the association between ctDNA and the prognosis of patients diagnosed with MM. A comprehensive literature search was performed across PubMed, Embase, We…
