The present study aimed at developing an original pre-column HPLC assay allowing rapid characterization of nitric oxide (NO) scavengers from complex plant extracts. Sodium nitroprusside (SNP) was employed as a NO donor and spiked with an aqueous extract from Aloysia triphylla leaves prior to HPLC analysis. Relying on the ability of radical scavenging constituents to be oxidized upon reaction with radicals, this assay successfully allowed direct identification of three potential NO scavengers, including verbascoside, isoverbascoside, and luteolin-7-O-diglucuronide. These three phenolics were also individually assessed for their NO scavenging activities by using a Griess colorimetric assay. With respective IC50 values of 56 ± 4, 51 ± 3, and 69 ± 5 µg/mL, verbascoside, isoverbascoside, and luteolin-7-O-diglucuronide were all reported as potent NO scavenging compounds, confirming the efficiency of the SNP spiking HPLC assay. The present method can, thus, be considered as a valuable and effective approach for speeding up the discovery of NO scavenging constituents.

Dietary supplements are commonly used by animals and humans and play key roles in diverse systems, such as the immune and reproductive systems, and in metabolism. Essential oils (EOs), which are natural substances, have potential for use in food supplementation; however, their effects on organisms remain to be elucidated. Here, we examine the effects of dietary Aloysia triphylla EO supplementation on zebrafish behavior, metabolism, stress response, and growth performance. We show that fish fed diets containing A. triphylla EO presented an anxiolytic response, with reduced exploratory activity and oxygen consumption; no changes were observed in neuroendocrine stress axis functioning and growth was not impaired. Taken together, these results suggest that the A. triphylla EO supplementation is a strong candidate for use in feed, since it ensures fish welfare (anxiolytic behavior) with decreased oxygen consumption. This makes it suitable for use in high-density production systems without causing damage to the neuroendocrine stress axis and without growth performance being impaired.

The objective of this study was to identify and quantify the chemical constituents, as well as the antimicrobial, antibiofilm and synergistic activity with florfenicol of essential oils of Aloysia triphylla (EOAT) and Lippia alba (EOLA) against Aeromonas spp. The antimicrobial activity of EOAT and EOLA was verified by the minimum bactericidal concentration (MBC) and the action against biofilm forming and consolidated biofilm. The synergistic activity of EOAT and EOLA with florfenicol was performed by the checkerboard technique. The main components of EOAT were α-citral (39.91%), E-carveol (25.36%) and limonene (21.52%), while that of EOLA was linalool (81.64%). Aeromonas spp. isolates showed sensitivity to both essential oils with MBC between 195.3 and 3125.0 μL/mL. Two isolates were classified as non-producing, three as moderate and 16 as weak biofilm producers. The EOAT and EOLA interfered in the biofilm formation, from moderate to weak producers, but did not cause any interference in the consolidated biofilm. The EOAT and EOLA combined with florfenicol showed synergistic effect and reduced MBC. The EOAT and EOLA have potential for application as antimicrobial agents, as they interfere in the initial formation of biofilm and when combined with florfenicol, present a synergic effect with a reduction in the minimum dose of the antibiotic.

Aloysia triphylla (Verbenaceae) is an aromatic medicinal plant, and it is used for the treatment of "nervous" problems as, "sadness" and "nervousness". While, there are no reports about its pharmacological activity in animal models. The objective of this work was to evaluate the anxiolytic effect of the extracts and fractions of this species and to measure the interaction of the most active fraction with serotonergic, glutamatergic and GABAergic drugs. An elevated plus maze test was carried ought where the methanol (AtM), dicloromethane (AtD) and hexanic (AtH) extracts presented anxiolytic activity in mice when exposed to the test. Also, different fractions obtained from the AtD were evaluated (AtF1, AtF2 and AtF3, 15mg/kg), and showed that fraction AtF1 possessed the anxiolytic activity, in the same model. Then, AtF1 was co-administered with different drugs, which act on GABAergic (bicuculline, picrotoxin, pentylenetetrazol, baclofen and phaclofen), or serotononinergic (DOI, 8-OH-DPAT, WAY 100635 and ketanserine) or glutamatergic (NMDA, MPEP and MK-801) systems. The anxiolytic activity of AtF1 was modified by GABAergic and serotoninergic drugs. Chemical analysis of this fraction by using GC-MS, showed that it contains hexadecanoic acid, hexadecanoic acid methyl ester, octadecanoic acid methyl ester, eicosanoic acid methyl ester, vitamin E, α-amiryn, campesterol, sitosterol, stigmastan-2,22, dien-3-ol (4) and stigmasta 5, 24 (28) dien-3-ol.

The essential oil of Aloysia triphylla (EOAT) is a promising product with potential use in aquaculture systems. This study evaluated hematological/biochemical responses and survival of silver catfish (Rhamdia quelen) fed a diet containing EOAT and infected by Aeromonas hydrophila. After 21 days of feeding trial, fish were infected with A. hydrophila following a 10-day period of observation. Blood collection was performed before and after the bacterial challenge. Dietary EOAT by itself seems to affect some blood parameters, decreasing total leukocyte, lymphocyte, and neutrophil counts and increasing total protein values. However, 2.0 mL EOAT/kg diet showed a possible potential protective effect after A. hydrophila infection, maintaining the evaluated parameters similar to basal values (from healthy fish before the feeding trial) and promoting survival of silver catfish.

Verbascoside (acteoside), a phenyl propanoid glycoside, comprises 0.5 to 3.5 % dry weight of Lippia citriodora leaves. A wide range of biological activities are attributed to verbascoside including anti-inflammatory, antioxidant, anti-bacterial, anti-tumor, anti-fungal, photoprotective as well as chelating effects. The objective of this study is to evaluate the effect of verbascoside on pregnancy outcome in mice. Timed-pregnant mice received doses of 1g/kg/day verbascoside or the vehicle control during organogenesis, intraperitoneally. Maternal body weights were measured throughout pregnancy. The litters were examined for external malformations and skeletal abnormalities. Then they were stained with Alizarin red S and Alcian blue. Maternal exposure to verbascoside throughout pregnancy did not influence the mean of maternal weight gain. Statistically significant difference was not found in mean number of implantation sites, live and resorbed fetuses between control and experiment groups. Our data demonstrate that the main component of L. citriodora, verbascoside using during organogenesis possesses no risk to fetuses. However, more research projects are needed to confirm these findings and determine the exact effects of verbascoside on human embryo development.

Essential oils (EOs) are widely used as biopesticides and to control bacterial infections. This study describes the ability of six EOs isolated from plants cultivated in Colombia to perform as repellents against Ulomoides dermestoides and as quorum sensing (QS) inhibitors. EOs from Aloysia triphylla, Cymbopogon nardus, Lippia origanoides, Hyptis suaveolens, Swinglea glutinosa and Eucalyptus globulus were repellents classified as Class IV, IV, IV, III, II, and II, respectively, whereas the commercial repellent IR3535 only reached Class II after 2 h exposure. All EOs presented small, but significant inhibitory properties against the QS system in Escherichia coli (pJBA132) at 25 μg/mL after 4 h exposure. These data suggest evaluated EOs from Colombia are sustainable, promising new sources of natural repellents and could be important as anti-quorum sensing molecules.

Eleven Algerian medicinal and aromatic plants (Aloysia triphylla, Apium graveolens, Coriandrum sativum, Laurus nobilis, Lavandula officinalis, Marrubium vulgare, Mentha spicata, Inula viscosa, Petroselinum crispum, Salvia officinalis, and Thymus vulgaris) were selected and their hydroalcoholic extracts were screened for their antiradical and antioxidant properties in cell-free systems. In order to identify the main metabolites constituting the extracts,1H NMR-based metabolic profiling was applied. Data obtained emphasized the antiradical properties of T. vulgaris, M. spicata and L. nobilis extracts (RACI 1.37, 0.97 and 0.93, respectively), whereas parsley was the less active as antioxidant (RACI -1.26). When the cytotoxic effects of low and antioxidant doses of each extract were evaluated towards SK-N-BE(2)C neuronal and HepG2 hepatic cell lines, it was observed that all the extracts weakly affected the metabolic redox activity of the tested cell lines. Overall, data strongly plead in favor of the use of these plants as potential food additives in replacement of synthetic compounds.
