- Palonosetron-induced anaphylaxis during general anesthesia in a patient with multiple drug hypersensitivity. [Journal Article]Int J Clin Pharmacol Ther. 2026 Aug 18. [Online ahead of print]IJ
- CONCLUSIONS: Prior uneventful exposure to palonosetron does not guarantee future safety. Atopic patients with polysensitization are at significantly increased risk of severe anaphylaxis upon re-exposure. Positive ondansetron skin testing provides indirect evidence identifying palonosetron as the culprit. All 5-HT3 receptor antagonists should permanently be avoided in such patients, and alternative antiemetics (e.g., dexamethasone, droperidol, NK1 antagonists) should be selected.

- Tislelizumab-induced hyperamylasemia: Case report and safety analysis using the FAERS database. [Journal Article]Int J Clin Pharmacol Ther. 2026 Aug 18. [Online ahead of print]IJ
- CONCLUSIONS: Tislelizumab is associated with multisystem adverse events. Close monitoring of serum amylase levels, as well as regular hematological and biochemical examinations, is essential during clinical medication.

- Clinical Significance of Acute Kidney Injury in Idiosyncratic Drug-Induced Liver Injury: A Multicentric Propensity Scores Matched Study. [Journal Article]
- Evidence about the role of acute kidney injury (AKI) in idiosyncratic drug-induced liver injury (DILI) is still scarce. We aimed to ascertain the incidence, clinical profile, culprit drugs, and prognosis associated with concomitant DILI and AKI. We include patients from two long-term prospective DILI registries, the Spanish DILI registry and the Latin American DILI (LATINDILI) Network. Demographi…

- Enzalutamide-Induced Host Metabolic Reprogramming Is Associated with Treatment Outcomes in Patients with Castration-Resistant Prostate Cancer: A Hypothesis-Generating Study. [Journal Article]
- Host metabolic changes in response to drug treatment may shape individual pharmacological outcomes. This study aims to identify serum metabolomics signatures potentially associated with individual responses to enzalutamide treatment in metastatic castrate-resistant prostate cancer (mCRPC) patients. Targeted metabolomic profiles in serum collected from 18 mCRPC patients treated with 160 mg/day of …

- Tramadol Co-Administration with Oxycodone or Hydrocodone Increases Opioid Burden without Improving Postoperative Pain Control. [Journal Article]
- Tramadol is frequently co-prescribed with oxycodone or hydrocodone after surgery under the assumption that its dual mechanism of action provides an opioid-sparing benefit. However, evidence supporting this practice is limited and inconsistent. We conducted a secondary analysis of the multicenter IGNITE ADOPT-PGx pragmatic trial of patients undergoing elective surgery who were prescribed oxycodone…

- Translational Paradox of Triplet Repeat Expansion Disorders: Synthesizing Clinical Trial Failures to Guide Future Therapeutics. [Review]
- The comprehensive understanding of triplet repeat expansion (TRE) disorders as monogenic neurodegenerative diseases presents a fundamental challenge for developing treatments capable of modifying the disease course, as neurogenetic research indicates. The past 30 years reveal that preclinical results demonstrating strong effectiveness have not translated into success in human clinical trials, wit…

- Triglyceride Polygenic Score Identifies Individuals Who May Respond Differently to Aspirin in Primary Prevention. [Journal Article]
- Low-dose aspirin is no longer routinely recommended for the primary prevention of cardiovascular disease in older adults due to a lack of net benefit over bleeding risk. We hypothesized that genetic subgroups may experience differential harm or benefit from aspirin therapy. To investigate this, we screened 572 polygenic scores (PGSs) for modification of aspirin's effect on major bleeding and majo…

- Predictors of anamorelin discontinuation in patients with cancer-associated cachexia undergoing chemotherapy: A retrospective observational study. [Journal Article]Int J Clin Pharmacol Ther. 2026 Aug 14. [Online ahead of print]IJ
- CONCLUSIONS: In patients with cancer-associated cachexia, baseline PS and PNI were associated with anamorelin discontinuation. Furthermore, competing risk analysis demonstrated PS was the primary determinant of anamorelin discontinuation, whereas PNI may be more closely associated with mortality risk. Therefore, early diagnosis of cancer-associated cachexia and timely initiation of anamorelin before declines in PS and PNI may enable long-term administration.

- Integrated assessment of rifampicin-mediated induction of transporters and drug-metabolizing enzymes in a long-term human liver tissue chip system. [Journal Article]
- Rifampicin is a prototypical clinical inducer used in drug-drug interaction (DDI) studies. However, the clinical relevance and predictability of rifampicin-mediated hepatic transporter induction remain poorly defined. Conventional hepatocyte culture models fail to capture clinically relevant regulation of transporters and drug-metabolizing enzymes (DMEs), particularly at the protein level. In thi…

- Population Pharmacokinetics and Exposure-Response Analyses to Support Cofetuzumab Pelidotin (ABBV-647) Dose Optimization in Non-Small Cell Lung Cancer Patients. [Journal Article]
- Cofetuzumab pelidotin (Cofe-P) is an anti-protein tyrosine kinase 7 (PTK7) antibody-drug conjugate (ADC) being investigated for the treatment of adults with PTK7-expressing, recurrent non-small cell lung cancer (NSCLC). The first-in-human (FIH) trial demonstrated tolerability of dosages ≤ 2.8 mg/kg once every 3 weeks (Q3W) and ≤ 3.2 mg/kg Q2W. Population pharmacokinetics (popPK) and exposure-resp…

- Regulatory Research Priorities for AI Use in the Medicine Lifecycle: A European Perspective with Global Relevance. [Journal Article]
- Regulatory bodies play a central role in providing guidance that enables safe and effective use of artificial intelligence tools in medicine development and evaluation. Regulators can also act as catalysts for regulatory science research. To inform these efforts, a European-wide survey was conducted to solicit stakeholder perspectives on the priority areas for regulatory science research related …
