- Maximal use of 0.05% topical isotretinoin in patients with congenital ichthyosis results in low systemic exposure. [Journal Article]Br J Clin Pharmacol. 2026 Apr; 92(4):1256-1260.BJ
- Congenital ichthyoses (CI) are rare, inherited skin disorders characterized by hyperkeratosis, scaling and fissuring that significantly impair patients' quality of life. Treatment options are limited, with systemic retinoids reserved for severe cases owing to their adverse effect profile. This open-label, single-arm, maximal-use trial investigated the systemic exposure and safety of a topically a…

- The history, development and current status of isotretinoin: a review article. [Journal Article]Clin Exp Dermatol. 2025 Dec 11. [Online ahead of print]CE
- Isotretinoin, a potent retinoid, is highly effective in the treatment of severe acne vulgaris. Its nearly 60-year history began with the Swiss pharmaceutical company La Roche in the 1960s, where it was initially studied for potential benefits in cancer treatment. This article delineates the historic trajectory of isotretinoin from early clinical trials in the 1970s and 1980s, highlighting its eff…

- Prescribing patterns for treatment of acne vulgaris: A retrospective chart review at an urban public and private hospital. [Multicenter Study]
- CONCLUSIONS: Findings from this study highlight differences in acne prescribing patterns by race/ethnicity and hospital system, which can impact the ability of patients to have successful treatment of their acne and its sequelae.

- Patterns of recurrence after radiotherapy for high-risk neuroblastoma: Implications for radiation dose and field. [Multicenter Study]
- CONCLUSIONS: Despite EBRT, LRR remains a contributor to treatment failure in HR-NBL with approximately half of LRRs including a component of marginal failure. Future prospective studies are needed to explore whether radiation fields and doses should be defined based on molecular features such as MYCN amplification, and/or response to chemotherapy.

- The Microbiome and Acne: Perspectives for Treatment. [Review]
- The skin microbiome consists of the microorganisms populating the human skin. Cutibacterium acnes (C. acnes, formerly named Propionibacterium acnes) is recognized as a key factor in acne development, regulating inflammatory and immune pathways. Dysbiosis has been described as the imbalance in skin microbiome homeostasis and may play a role in acne pathogenesis. Microbial interference has been sho…

- Effect of isotretinoin on CYP2D6 and CYP3A activity in patients with severe acne. [Journal Article]
- CONCLUSIONS: Administration of isotretinoin did not significantly reduce CYP2D6 activity in extensive metabolizers, suggesting that the predicted downregulation of CYP2D6 based on in vitro data does not translate into humans. We observed a modest increase in CYP3A activity (predominantly CYP3A4) with isotretinoin treatment. The data also suggest that DX glucuronidation is increased following isotretinoin administration.

- Compliance with Pregnancy Prevention Recommendations for Isotretinoin Following the Amendment of the European Union Pregnancy Prevention Program: A Repeat Study in Estonia. [Journal Article]
- CONCLUSIONS: A slight improvement in complete contraceptive coverage during isotretinoin use has not resulted in a decrease in the risk of isotretinoin-exposed pregnancies. The contraceptive usage and risk for pregnancy vary greatly across age groups, suggesting the need for a more targeted approach to improve the effectiveness of the PPP.

- Topical Isotretinoin (TMB-001) Treatment for 12 Weeks Did Not Result in Clinically Relevant Laboratory Abnormalities in Participants with Congenital Ichthyosis in the Phase 2b CONTROL Study. [Journal Article]
- CONCLUSIONS: There were no clinically significant laboratory changes in participants receiving TMB-001 isotretinoin ointment through 12 weeks of treatment, which differs from reported results for systemic isotretinoin.

- Phase IIb randomized CONTROL study demonstrates a novel topical isotretinoin formulation, TMB-001, is safe and effective in participants with either recessive X-linked or autosomal recessive lamellar congenital ichthyosis. [Randomized Controlled Trial]
- CONCLUSIONS: Regardless of congenital ichthyosis subtype, TMB-001 demonstrated greater proportions of participants achieving VIIS 50 and ≥ 2-grade IGA improvement vs. vehicle.

- Drug Survival of Oral Retinoids in Hidradenitis Suppurativa: A Real-Life Cohort Study. [Journal Article]
- CONCLUSIONS: Comparable drug survival rates at 12 months for isotretinoin and acitretin were found. HS patients with widespread comedones and the scarring folliculitis phenotype could benefit from treatment with isotretinoin or acitretin, respectively.

- Dermatology: how to manage acne in skin of colour. [Review]
- Acne vulgaris is a prevalent dermatological condition worldwide but is especially challenging to treat in individuals with skin of colour (SOC). Corresponding to Fitzpatrick skin phototypes III-VI, people of African, Asian, Middle Eastern and Hispanic ethnicity are considered to have SOC. With the additional risk of postinflammatory hyperpigmentation (PIH) as a consequence of inflammatory acne or…

- The road to biologics in patients with hidradenitis suppurativa: a nationwide drug utilization study. [Journal Article]
- CONCLUSIONS: Patients with HS who receive biologic therapy have long preceding treatment histories with multiple drug classes and treatment series, many of which are supported by relatively weak evidence in HS. Delay in the initiation of biologic therapy may represent a missed opportunity to prevent disease progression. What is already known about this topic? The treatment journey leading to biologic therapy in patients with HS has not previously been investigated. What does this study add? Our data from 225 patients with HS illustrate that patients who receive biologic therapy have long preceding treatment histories with multiple drug classes and treatment series, many of which are supported by relatively weak evidence in HS.

- Therapeutic Options for the Treatment of Darier's Disease: A Comprehensive Review of the Literature. [Review]
- Darier's disease (also known as keratosis follicularis or dyskeratosis follicularis) is an autosomal dominant inherited disorder which manifests as hyperkeratotic greasy papules in the first or second decade of life. Aside from symptom management and behavioral modifications to avoid triggers, there are currently no validated treatments for Darier's disease (DD). However, a variety of treatments …
