- Adipose Tissue Dysfunction in Metabolic Dysfunction-Associated Steatotic Liver Disease. [Review]Cells. 2026 Jul 30; 15(15).C
- Metabolic dysfunction-associated steatotic liver disease (MASLD) is clinically heterogeneous. Patients with similar body mass index, waist circumference, or noninvasive fibrosis stratum may differ markedly in histologic activity, fibrosis trajectory, and progression risk. Adipose tissue dysfunction helps explain this variation. Adipose dysfunction has been characterized by direct depot quantifica…

- Association of pioglitazone versus dipeptidyl peptidase-4 inhibitor use with incident gout in type 2 diabetes: a real-world cohort study. [Journal Article]Acta Clin Belg. 2026 Aug 10; :1-8. [Online ahead of print]AC
- CONCLUSIONS: Pioglitazone use is associated with a modestly higher incidence of gout compared with DPP-4i use in patients with T2DM; however, the small absolute risk difference and observational design warrant cautious interpretation.

- The new era of MASH pharmacotherapy: a comprehensive review of FDA-approved and emerging agents. [Review]Front Gastroenterol (Lausanne). 2026; 5:1850915.FG
- CONCLUSIONS: The approval of resmetirom and semaglutide marks a new era in MASH management. A risk-stratified algorithmic approach using NITs guides patient selection, with liver biopsy reserved for specific clinical scenarios. The therapeutic landscape is rapidly evolving, with saroglitazar and combination approaches representing key frontiers.

- Effects of dietary restriction, methionine restriction, and pioglitazone on metabolic profile, blood glucose regulation, and bone quality in male middle-aged Sprague-Dawley rats. [Journal Article]Nutrition. 2026 Aug 06; 151:113353. [Online ahead of print]N
- CONCLUSIONS: While all interventions improved energy metabolism, DR and MR resulted in a minor compromise in bone mineral regulation relative to pioglitazone treatment in male SD rats.

- Dapagliflozin & pioglitazone combination therapy in T2DM with or without MASLD - a systematic review and meta-analysis: PRO-2 study. [Systematic Review]Front Clin Diabetes Healthc. 2026; 7:1733995.FC
- CONCLUSIONS: Dapagliflozin-pioglitazone combination therapy was associated with improvements in glycemic control and non-invasive markers of hepatic steatosis in adults with T2DM, with or without MASLD, while maintaining a favorable safety profile. Certainty of evidence was moderate for metabolic and steatosis outcomes and low for histological endpoints. Given the predominance of surrogate hepatic endpoints and limited biopsy-confirmed data, these findings should be interpreted with appropriate caution.

- Efficacy and safety of pioglitazone in lichen planopilaris in comparison with hydroxychloroquine: Prospective non-inferiority study. [Journal Article]J Dtsch Dermatol Ges. 2026 Aug 02. [Online ahead of print]JD
- CONCLUSIONS: This study provides the largest prospective pioglitazone cohort reported to date in LPP and demonstrates that pioglitazone is non-inferior to hydroxychloroquine as a first-line systemic treatment, while also exhibiting a strong safety profile.

- Fibrocalculous Pancreatic Diabetes: Changing Clinical Spectrum and Glycaemic Response to Pioglitazone. [Journal Article]Indian J Endocrinol Metab. 2026 May-Jun; 30(3):223-227.IJ
- CONCLUSIONS: FCPD in North Karnataka demonstrated atypical clinical features (low BMI and chronic diarrhoea) with delayed diagnosis and frequent hypoglycaemia. Glycaemic control could be optimised with short-acting insulin. Pioglitazone compared with metformin significantly improved glycaemic parameters and reduced insulin requirement, due to its effect on hepatic insulin resistance.

- Integrated computational and in vivo antidiabetic evaluation of benzylidene thiazolidinedione compounds. [Journal Article]RSC Med Chem. 2026 Jul 17. [Online ahead of print]RM
- Diabetes mellitus (DM) remains a major global health challenge, highlighting the need for safer and more effective antidiabetic therapies. In the present study, a series of acetyl pyrrolidine-2-carbonitrile-based benzylidene-thiazolidine-2,4-dione hybrids (5a-5i) were designed, synthesized and structurally characterized using standard spectroscopic techniques. The synthesized compounds were evalu…

- CONCLUSIONS: SGLT2i, GLP1RA, tirzepatide and (with lower quality of evidence) pioglitazone and metformin are associated with a reduced incidence of cardiovascular events; tirzepatide, SGLT2i and GLP1RA are also associated with a reduced all-cause mortality.

- Neurodegenerative diseases' treatments still represent one of the major unmet medical needs, and their related clinical trials are characterized by one of the highest failure rates. Despite the promising preclinical premises, pioglitazone lies among those drugs that failed clinical translation. It is an antidiabetic drug acting as a PPARγ agonist and later repurposed as a competitive and selectiv…

- Clinical Experience with Quintuple Non-Insulin Drug Therapy in Suboptimally Controlled Type 2 Diabetes. [Journal Article]Curr Drug Saf. 2026 Jul 25. [Online ahead of print]CD
- CONCLUSIONS: The prevalence of individuals with T2D treated with quintuple non-insulin drug regimens was 2.7% without any notable side effects or safety concerns. In patients who refuse insulin therapy, these combinations can be taken into consideration according to the characteristics of every individual and carefully evaluated for their safety and effectiveness.

- SERCA2 gatekeeper role in aortic autophagy: targeting the Ca[2+]-mTOR axis to prevent aortic dissection. [Journal Article]
- Aortic dissection (AD) is a catastrophic cardiovascular syndrome with an in-hospital mortality of more than 90%. We previously identified oxidative inactivation of sarcoplasmic/endoplasmic reticulum Ca[2+]-ATPase 2 (SERCA2) at cysteine 674 (C674) as a driver of aortic smooth muscle cell (ASMC) phenotypic switching. However, its causal impact on autophagic flux and AD remains unresolved. SERCA2 C6…

- Clinical efficacy of liraglutide combined with pioglitazone in obese polycystic ovary syndrome and its effects on the serum chemerin/visfatin ratio. [Journal Article]Am J Transl Res. 2026; 18(6):5128-5144.AJ
- CONCLUSIONS: Liraglutide combined with pioglitazone can effectively ameliorate body mass, glucose and lipid metabolism, reproductive hormone levels and IR in obese PCOS patients, without increasing the risk of adverse events. The serum chemerin/visfatin ratio is closely correlated with IR and may serve as a promising biomarker for predicting treatment response in obese PCOS.
