- Itch Relief and Quality-of-Life Improvement with Abrocitinib and Dupilumab in Patients with Moderate-to-Severe Atopic Dermatitis: A Post Hoc Analysis of JADE COMPARE and JADE DARE. [Journal Article]
- CONCLUSIONS: In this post hoc pooled analysis of the JADE COMPARE and JADE DARE trials, the proportion of patients with moderate-to-severe atopic dermatitis achieving clinically meaningful itch responses, including an itch-free state, was generally higher with abrocitinib compared with dupilumab. Median time to achievement of both PP-NRS4 and an itch-free state was shorter with abrocitinib than with dupilumab. Patients who achieved an itch-free state experienced greater improvements in quality of life and sleep compared with patients with residual itch.

- CONCLUSIONS: A variety of demographic, clinical, and biomarker-related factors contribute to the development of cirAEs. Characterizing these risk factors can address clinical needs for cirAE identification, while further mechanistic studies are needed to enhance management strategies. However, there is a limited amount of high-quality prospective evidence on these risk factors, and the quality of reporting on immunotherapy-related adverse events is inconsistent. Future research should focus on validating clinically valuable risk factors and interrogating mechanisms underlying cirAEs emergence.

- CONCLUSIONS: This report of upadacitinib in SAPHO syndrome demonstrates its rapid and substantial efficacy in a patient refractory to adalimumab. Upadacitinib may represent a promising treatment option for difficult-to-treat SAPHO syndrome, particularly in cases with inadequate response to TNF-α inhibition.
