- Overcoming Immunotolerance in Chronic Hepatitis B: Efficacy of Granulocyte-Macrophage Colony-Stimulating Factor-Based Immunotherapy in Achieving Hepatitis B Surface Antigen Seroclearance. [Journal Article]MedComm (2020). 2026 Apr; 7(4):e70676.M
- Chronic hepatitis B (CHB) remains incurable due to the immune system's tolerance toward the hepatitis B virus (HBV) surface antigen (HBsAg). This study aimed to achieve a functional cure by breaking HBV tolerance through immunotherapy. CHB patients were treated with either standard nucleotide analog (NA) therapy (Adefovir Dipivoxil, ADV) (Cohort 1) or ADV combined with interferon-alpha (IFN-α) (C…

- Adefovir dipivoxil-induced hypophosphatemic osteomalacia and osteoporosis: a case report and literature review. [Case Reports]Front Endocrinol (Lausanne). 2026; 17:1766477.FE
- The nephrotoxicity of adefovir dipivoxil (ADV) can induce Fanconi syndrome, which causes hypophosphatemic osteomalacia. We report a case of a 67-year-old postmenopausal woman who had been receiving long-term adefovir dipivoxil therapy for chronic hepatitis B. She presented with severe bone pain and multiple pseudofractures, and was diagnosed with drug-induced hypophosphatemic osteomalacia. Follow…

- Hypophosphatemic Osteomalacia Induced by Low-Dose Adefovir-Related Fanconi Syndrome. [Case Reports]
- Adefovir dipivoxil (ADV), an antiviral agent for chronic hepatitis B virus infection, has been associated with nephrotoxicity, particularly Fanconi syndrome, even at low doses (10 mg/day). Fanconi syndrome is a generalized proximal tubular dysfunction leading to phosphate wasting and hypophosphatemic osteomalacia. Here, the author reports a case of a 58-year-old man with a 2-year history of bone …

- Adefovir dipivoxil-induced Fanconi syndrome associated with hypophosphatemic osteomalacia misdiagnosed as cervical spondylotic myelopathy: A case report and literature review. [Case Reports]
- CONCLUSIONS: Fanconi syndrome with hypophosphatemic osteomalacia induced by low-dose adefovir dipivoxil (10 mg/d) is extremely rare. The condition may present with pain, muscle weakness, or numbness, and its nonspecific manifestations can easily lead to a misdiagnosis of cervical spondylotic myelopathy. For patients receiving long-term ADV therapy who develop such symptoms, especially in the presence of cervical disc herniation with spinal cord compression, clinicians should carefully evaluate laboratory findings and physical examination results to exclude the possibility of ADV-induced Fanconi syndrome.

- Monkeypox Virus Is Inhibited by Nucleoside Analogues Including the Acyclic Phosphonates Adefovir and Tenofovir In Vitro. [Journal Article]J Med Virol. 2025 Nov; 97(11):e70669.JM
- Mpox, caused by the orthopoxvirus monkeypox virus (MPXV), has garnered worldwide attention during a global outbreak in 2022 and in an ongoing outbreak in Central Africa. Because of drug resistance and toxicity risks with current antiviral treatments, there is still a need for additional antivirals to treat mpox and other orthopoxvirus diseases. We developed in vitro screening assays using recombi…

- Human Embryoid Body-Based Assays for Preclinical Screening of Potential Teratogenic Drugs: Antiviral Drugs for Hepatitis B Virus as Examples. [Journal Article]Stem Cells Dev. 2025 Oct; 34(19-20):427-439.SC
- Ensuring the safety of drugs during pregnancy is a critical concern in clinical practice. Animal tests are not always reliable for accurately identifying human teratogens, as evidenced by the infamous thalidomide case. Therefore, there is an urgent need for rapid, dependable, and cost-effective human in vitro assays for potential teratogenic drugs. Here, we utilized human embryoid body (hEB)-base…

- Adefovir dipivoxil-associated Fanconi syndrome combined with peripheral neuropathy: a case report and literature review. [Journal Article]
- Long-term use of low-dose adefovir dipivoxil (ADV) has been increasingly associated with Fanconi syndrome; however, cases of Fanconi syndrome combined with peripheral neuropathy remain rare. Here, we report a 67-year-old man who developed progressive limb weakness and sensory abnormalities after approximately 6 years of treatment with 10 mg ADV. He was diagnosed with ADV-associated Fanconi syndro…

- QuEChERS and UPLC-MS/MS-Based Quantification of Human Plasma of Eight Nucleoside Reverse Transcriptase Inhibitors and Platinum Anticancer Drugs for Hepatocellular Carcinoma. [Journal Article]
- Nucleoside reverse transcriptase inhibitors (NRTIs) and platinum-based chemotherapeutics are widely utilized in cancer treatment. Evidence suggests that drug plasma concentrations are closely linked to both therapeutic efficacy and the risk of adverse effects. Consequently, developing therapeutic drug monitoring (TDM) methods is essential. Here, an effective procedure utilizing QuEChERS (Quick, E…

- FDA-approved drug repurposing screen identifies inhibitors of SARS-CoV-2 pseudovirus entry. [Journal Article]
- CONCLUSIONS: We identified four novel FDA-approved candidate drugs for anti-SARS-CoV-2 combination therapy. Our findings contribute to the growing body of evidence supporting drug repurposing as a viable strategy for rapidly developing COVID-19 treatments.

- Evaluation of Adefovir PBPK Model to Assess Biomarker-Informed OAT1 Drug-Drug Interaction and Effect of Chronic Kidney Disease. [Journal Article]
- Evaluation of transporter-mediated drug-drug interactions (DDI) with endogenous biomarkers, coupled with physiologically-based pharmacokinetic modeling (PBPK) is envisioned to replace or reduce dedicated DDI clinical trials with clinical probe substrates. The current study developed a PBPK model of adefovir (OAT1 clinical probe) by incorporating experimental measurements of adefovir passive diffu…

- Clinical management of hypophosphatemic osteomalacia induced by adefovir and tenofovir: Insights from a case report. [Case Reports]
- CONCLUSIONS: Accurate diagnosis requires a combination of clinical presentation, medical history, biochemical and radiological findings, and, if available, measurement of fibroblast growth factor 23 (FGF 23). The role of national, provincial, or regional centers for rare diseases is crucial for conducting unconventional tests and providing access to rare medications.

- Unachieved antiviral strategies with acyclic nucleoside phosphonates: Dedicated to the memory of dr. Salvatore "Sam" Joseph Enna. [Review]
- Many acyclic nucleoside phosphonates such as cidofovir, adefovir dipivoxil, tenofovir disoproxil fumarate, and tenofovir alafenamide have been marketed for the treatment or prophylaxis of infectious diseases. Here, this review highlights potent acyclic nucleoside phosphonates for their potential in the treatment of retrovirus (e.g., human immunodeficiency virus) and DNA virus (e.g., adeno-, papil…

- Understanding adefovir pharmacokinetics as a component of a transporter phenotyping cocktail. [Journal Article]
- CONCLUSIONS: The popPK modeling approach indicated that the co-administration primarily affected the apparent absorption and/or prodrug conversion of adefovir dipivoxil, resulting in the minor drug-drug interaction observed for adefovir as a victim. However, renal elimination remained unaffected. The high Km value suggests that assessing renal OAT1 activity by CLR has no relevant misspecification error with the cocktail doses used.
